竹節(jié)參化學(xué)成分研究

竹節(jié)參為五加科植物竹節(jié)參Panax japonicus C.A.Mey.的干燥根莖[1],又名竹節(jié)三七、竹節(jié)人參、白三七,主要分布于長江以南,西起云南西部,東至日本,且存在狹葉竹節(jié)參、珠子參等變種。竹節(jié)參具有滋補強壯、散瘀止痛、止血袪痰的功能,主要用于病后虛弱、勞嗽咯血、咳嗽痰多、跌撲損傷等癥[1],對中樞神經(jīng)系統(tǒng)、免疫系統(tǒng)、心血管系統(tǒng)具有藥理作用[2]。竹節(jié)參的主要活性成分為皂苷類,并含少量揮發(fā)油、多糖和氨基酸等[2]。國內(nèi)學(xué)者對竹節(jié)參的化學(xué)成分研究主要集中在三萜皂苷類化合物,而很少報道過嘌呤嘧啶類化合物,我們對竹節(jié)參進行了化學(xué)成分研究,分離鑒定了12個化合物:竹節(jié)參皂苷Ⅳa(1)、楤木皂苷A(2)、竹節(jié)參皂苷Ⅰb(3)、竹節(jié)參皂苷Ⅴ(4)、尿嘧啶(5)、胸嘧啶(6)、尿苷(7)、鳥苷(8)、胸苷(9)、腺苷(10)、肌苷(11)、胞苷(12),其中5~12為首次從該植物中分離。

1 儀器與材料

Bruker AM-600核磁共振光譜儀,ESI-MS(MDS Sciex Qstar)型質(zhì)譜儀,BUCHI旋轉(zhuǎn)蒸發(fā)儀,大孔吸附樹脂(D101,天津市海光化工有限公司),MCI (CHP20P75-150 μm,Mitsubishi化工廠),葡聚糖凝膠Sephadex LH-20(美國Pharmacia公司),ODS(西安交大寶賽生物技術(shù)公司)柱層析硅膠(200~300目,青島海浪硅膠干燥劑廠),高效薄層色譜層析板(青島海洋化工廠)。

竹節(jié)參購自湖北恩施州中藥材研究所竹節(jié)參種植基地,經(jīng)武漢工業(yè)學(xué)院陳新教授鑒定為五加科植物竹節(jié)參Panax japonicas C.A.Mey的干燥根莖。

2 提取分離

干燥竹節(jié)參根莖5 kg,切片,粉碎,用90%乙醇25 L冷浸提取三次,每次一周,三次濾液分別濃縮后合并。依次用乙酸乙酯、正丁醇分別萃取三次,濃縮得到乙酸乙酯部位186 g,正丁醇部位208 g。乙酸乙酯部位和正丁醇部位合并一起上D101大孔樹脂柱,乙醇-水溶液梯度洗脫,收集并濃縮得到兩部分分別為Fr1、Fr2。Fr2上MCI柱,和反復(fù)硅膠柱色譜(三氯甲烷-甲醇-水 =4∶1∶20 μL-1∶3∶20 μL)得Fr3-2-1~6個部分,F(xiàn)r3-2-1用溶劑系統(tǒng)甲醇-水重結(jié)晶2次,得化合物1(366 mg)。Fr3-2-3,上Sephadex LH-20柱用甲醇洗脫,得化合物2(678 mg)和5(211 mg)。Fr3-3-2上ODS柱用溶劑系統(tǒng)乙腈-水梯度洗脫,得到化合物3(1.856 g)、9(110 mg)和12(98 mg)。Fr3-3-3分別用溶劑系統(tǒng)乙腈-H2O梯度洗脫,得到化合物4(208mg)和10(60 mg)。將Fr1上MCI柱,和反復(fù)硅膠柱色譜(三氯甲烷-甲醇=20∶1~1∶1)得到流分 Fr4-1-1、Fr4-1-2、Fr4-2-1和 Fr4-3-14個部分。Fr4-1-1上Sephadex LH-20柱用甲醇洗脫,得到化合物6(20 mg)。Fr4-1-2、Fr4-2-1和Fr4-3-1分別上 ODS柱得到化合物7(5 mg)、8(10 mg)和11(15 mg)。

3 結(jié)構(gòu)鑒定

4 結(jié)果討論

從干燥竹節(jié)參根莖中分離得到12個化合物,其中三萜皂苷類4個,嘌呤嘧啶類8個。研究表明竹節(jié)參總皂苷具有一定的抗氧化活性[8],可顯著提高運動后小鼠肝臟、腦組織中超氧化物歧化酶(SOD)和谷胱甘肽(CSH)活性,降低丙二醛(MDA)含量[8]。大量實驗研究結(jié)果表明嘌呤和嘧啶類化合物具有廣泛的特異性作用[9],如短時程作用方面(神經(jīng)信號傳遞、外分泌、內(nèi)分泌和免疫功能調(diào)節(jié)),以及長時程作用方面(發(fā)育及再生過程中的細胞生長、分化和增生)[9]。
