摘要 通過殼聚糖及戊二醛將雙胺氧化酶固定在普魯士藍修飾的玻碳電極表面,制備了生物胺傳感器.考察了該傳感器的電化學(xué)特性并優(yōu)化了實驗條件.結(jié)果表明,在優(yōu)化條件下所制備的傳感器的電流響應(yīng)與組胺濃度在0.33 μmol/L~3.3 mmol/L(3.7×10-2~3.7×102mg/L)范圍內(nèi)相關(guān);檢測限(S/N=3)為0.05 μmol/L (6 μg/L).該傳感器具有響應(yīng)范圍寬、靈敏度高、選擇性強、簡便快速的特點.

生物胺是一類具有生物活性的低分子量有機含氮化合物的總稱,主要通過氨基酸的脫羧作用生成[12].根據(jù)結(jié)構(gòu)可將其分為3類[3]:脂肪族(腐胺、尸胺、精胺、亞精胺等);芳香族(酪胺、苯乙胺等);雜環(huán)族(組胺、色胺等).適量的生物胺有助于人體的正常生理功能,過量則會引起不良的生理反應(yīng),如出現(xiàn)頭痛、心悸、腹瀉、休克等中毒癥狀[4].生物胺廣泛存在于肉類、水產(chǎn)品、乳制品、酒類和飲料等食品中,在食品中的含量與食品的質(zhì)量有一定的相關(guān)性[56].美國FDA規(guī)定水產(chǎn)品中組胺含量不超過50 mg/kg,酪胺不超過100 mg/kg,總胺不超過1 000 mg/kg;歐盟規(guī)定鯖科魚類中組胺含量不超過100 mg/kg,總胺不超過300 mg/kg[7].
檢測食品中的生物胺有助于監(jiān)控食品的質(zhì)量及安全性.現(xiàn)有的生物胺檢測方法主要有高效液相色譜法、氣相色譜法、薄層色譜法、毛細管電泳法等,這些方法存在費時、儀器昂貴、操作繁瑣等缺點.本文工作旨在研究開發(fā)靈敏、簡便、快速的電化學(xué)生物胺傳感器檢測方法.已有的這類傳感器大多直接檢測胺氧化酶與生物胺反應(yīng)生成的H2O2的氧化電流[8],或利用固定化辣根過氧化物酶和游離的電子媒介物檢測酶反應(yīng)生成的H2O2[910].前者工作電位較高,干擾較多;后者構(gòu)造相對3結(jié)論
采用殼聚糖和戊二醛固定化雙胺氧化酶,并利用普魯士藍對過氧化氫的高效電催化,制備了基于普魯士藍/雙胺氧化酶修飾電極的生物胺傳感器.該傳感器具有檢測范圍寬、檢測限低、選擇性好等優(yōu)點.有望為食品中生物胺的檢測提供一種簡便快速的方法.
