The drug-containing culture medium method for the test of toxicity was adopted to compare inhibitive effects of original nano-Cu2O drug and nano-Cu2O suspension,and nano-Cu2O drug has better inhibitive effects on snake melon Botrytis cinerea than original nano-Cu2O drug with the same mass concentration,and inhibitory effects are positively correlated with concentration.Correlation coefficients of the toxicity regression equation are 0.892 2 and 0.996 1,effective concentration EC50of original nano-Cu2O drug and that of nano-Cu2O suspension are 3 948.9 mg/ kg and 167.9 mg/kg.Original nano-Cu2O drug has an inhibitive effect on snake melon Botrytis cinerea,but the inhibition of nano-Cu2O suspension is more obvious.

S nake melon is one of the major vegetable species in the sunlight greenhouse in winter in Qinghai,while the snake melon botrytis cinerea produce negative effects on the snake melon.According to the research,the snake melon Botrytis cinerea can be very worse,and can lead to a reduction of 20%to 40%,sometimes even above 60%,which brings great loss to the production of snake melon[1-2].The overuse of certain chemical medicine can easily develop resistance to drugs.Only by developing new germicides and using different germicides in turn can we improve the potency of drugs and ease the resistance to drugs[3-4].Nano-Cu2O drug is a new visible P type semiconductor that serves as an absorbent,so there are already reports that Nano materials and technologies have been applied in the prevention of dieases[5-8]. The experiment used to determine the drug potency,and compares the inhibitory effects of Nano-Cu2O drug and Nano-Cu2O drug suspension on snake melon Botrytis cinerea.

Materials and methods

Tested fungi

Snake melon Botrytis cinerea was provided by Academy of Agriculture and Forestry Sciences of Qinghai University and had been tested.

Tested materials

The molecular weight of Cu2O was 143.09 and its average particle diameter was between 20 and 30 nm,which was purchased from Nanjing Guanye Chemical Industry in 2012. The molecular weight of CH3(CH2)15N(CH3)3Br was 364.45,and it was bought from Xiningmeida Co.,Ltd in 2012.

Testing methods

(1)Preparation of suspension solution.Nano-Cu2O and CH3(CH2)15N(CH3)3were added into the deionized water at the proportion of 1∶0.5,and emulsification lasted for 15 min,before mixing by the HJ-6A blender produced by Changzhou Guohua Electronic Equipment Ltd.,CO.at the maximum rotation speed of 1 400 r/min.The blender was set at the high position for 60 min,and the Nano-Cu2O suspension of different concentration were prepared.

(2)Preparation of medical culture medium.PDA culture medium was prepared based on Fang Zhongda’s method at the proportion of 1:9 of suspension solution and culture medium. There were six treatments,as the Nano-Cu2O in culture medium being 500,400,300,200 and 100 mg/kg. Snake melon Botrytis cinerea was beat into the fungi cake with a diameter of 3 mm fore use.The well-prepared culture medium was put into a flask for sterilization for 30 minutes. When the temperature fell to 50℃,the fungi cake was put into the plate before sending to the SPX-250-GB intelligent sunlight culture box for preparation at the temperature of 25℃.

(3)Data collection and statistics. The crossing method was applied on the fifth day to measure the diameter of fungi.Then the diameter of fungi growth was calculated according to the method by deducting fungi cake diameter from fungi diameter.The inhibitory effects of different concentration of nano-Cu2O drug were measured based on Mu Liyi[10],and then the drug potency regression and related coefficients were obtained by taking the effective component concentration as the independent variable,and inhibitory effects as dependent variables.After that,the EC50was calculated when the y=5 in the drug potency regression.

Inhibitory rate= [(Control fungi growth diameter- Disposed fungi growth diameter/Control fungi growth diameter)×100%.

Results and Analysis

Inhibitory effects of original nano-Cu2O drug on Botrytis cinerea

Nano-Cu2O drug was directly put into the culture medium.The inhibitory rates of original nano-Cu2O drug on Botrytis cinerea was 3.51%,7.02%,10.53%,12.28%and 15.79%when the quality concentration of nano-Cu2O drug was put into the culture medium directly(Table 1).

As can be seen from the drug potency regression formula in the Table 1,y=0.914 4+1.136x.When y=5,x= 3.596 5,the effective concentration EC50was 3 948.9 mg/kg.The inhibitory effects of original nano-Cu2O drug on Botrytis cinerea were active,but not significant.

Inhibitory effects of nano-Cu2O suspension on Botrytis cinerea

The inhibitory rates of nano-Cu2O suspension on Botrytis cinerea were 15.79%,75.44%,82.46%,87.72%and 89.47%,when the quality concentration of nano-Cu2O suspension on Botrytis cinerea was 100,200,300,400 and 500 mg/kg by putting nano-Cu2O suspension into the culture medium(Table 2).

According to the drug potency regression y=-2.023 9+3.156 6x in Table 2,when y=5,x=2.225 1,the effective concentration EC50was 167.9 mg/kg.The inhibitory effects of original nano-Cu2O suspension were significant,and the inhibitory effects showed positive correlation to the quality concentration of nano-Cu2O particles.

Conclusions

Nano-Cu2O drug has better inhibitive effects on snake melon Botrytis cinerea than original nano-Cu2O drug with the same mass concentration,and inhibitory effects are positively correlated with concentration.Correlation coefficients of the toxicity regression equation are 0.892 2 and 0.996 1,effective concentration EC50of original nano-Cu2O drug and that of nano-Cu2O suspension are 3948.9 mg/kg and167.9mg/kg.Original nano-Cu2O drug has an inhibitive effect on snake melon Botrytis cinerea,but the inhibition of nano-Cu2O suspension is more obvious.

納米Cu2O原藥與懸浮液對菜瓜灰霉病菌的抑制作用

為比較納米Cu2O原藥與懸浮液對菜瓜灰霉病菌的抑制作用,研究采用含藥培養(yǎng)基法進(jìn)行了毒力測定。試驗結(jié)果表明:同一質(zhì)量濃度的納米Cu2O懸浮液對菜瓜灰霉病菌的抑菌效果明顯優(yōu)于納米Cu2O原藥,抑制作用與質(zhì)量濃度呈正相關(guān);毒力回歸方程的相關(guān)系數(shù)分別為0.892 2和0.996 1,納米Cu2O原藥與懸浮液對菜瓜灰霉病菌的有效中濃度EC50分別為 3 948.9和 167.9 mg/kg.因此納米Cu2O原藥對菜瓜灰霉病菌有一定的抑制作用,而納米Cu2O懸浮液對菜瓜灰霉病菌的抑制作用非常明顯。
