紅花(Carthami Flos,CF)為菊科(Compositae)紅花屬(CarthamusLinn.)植物紅花(CarthamustinctoriusLinn.)的干燥管狀花[1]。紅花在漢代傳入中國(guó)中原地區(qū),最初只作為染料,之后主要作為中藥[2]。紅花作為染料主要用于染制絲織品和紙張,或者制作胭脂和口紅等化妝品[3]。紅花具有活血通經(jīng)和散瘀止痛之療效,臨床上主要用于治療經(jīng)閉、痛經(jīng)、癥瘕痞塊和跌撲損傷等,是一種常用的活血化瘀類中藥[4]?,F(xiàn)代藥理研究表明,紅花具有抗凝血、抗血栓形成、擴(kuò)張血管、改善微循環(huán)以及調(diào)節(jié)免疫等作用[5]7-11。目前,從紅花中已分離并報(bào)道醌式查爾酮苷類、黃酮類、亞精胺類、生物堿類、有機(jī)酸類和甾體類等200多種化合物[5]21。

紅花不僅應(yīng)用范圍廣、經(jīng)濟(jì)價(jià)值較高,而且環(huán)境適應(yīng)能力強(qiáng),世界各地都有栽培[6-7],中國(guó)新疆、河南、浙江、云南、四川和甘肅等地均有種植[2]。目前,紅花在中國(guó)的商業(yè)化種植主要集中在新疆、云南和甘肅,其中,在新疆的種植面積占中國(guó)紅花種植面積的50%以上,其紅花產(chǎn)量占中國(guó)紅花產(chǎn)量的80%以上[8]。紅花作為大宗中藥材,是否存在重金屬及有害元素含量超標(biāo)的問(wèn)題仍不明確,而《中國(guó)藥典》[1]僅對(duì)其有效成分的含量進(jìn)行限定,缺乏對(duì)紅花中重金屬及有害元素的檢測(cè)標(biāo)準(zhǔn)。紅花藥材的安全性影響其組成復(fù)方和注射液等產(chǎn)品的療效及安全性[9-10],因此,亟需加強(qiáng)對(duì)其無(wú)機(jī)元素組成規(guī)律的探索,提高用藥安全性。研究表明:土壤中無(wú)機(jī)元素的組成與植物體內(nèi)無(wú)機(jī)元素的含量有一定相關(guān)性,不同地區(qū)的土壤也會(huì)影響紅花作為藥材的安全性[11-12]。目前,國(guó)內(nèi)對(duì)紅花中無(wú)機(jī)元素組成和含量的研究大多集中于新疆產(chǎn)地,而針對(duì)其他產(chǎn)地紅花的研究不足,此外,還缺乏針對(duì)不同產(chǎn)地紅花重金屬及其他無(wú)機(jī)元素含量的對(duì)比研究。近期的研究結(jié)果表明:紅花生長(zhǎng)過(guò)程中對(duì)土壤中元素的吸收存在一定規(guī)律[13-14],因此,探究不同產(chǎn)地紅花中無(wú)機(jī)元素含量的分布規(guī)律具有一定的實(shí)際意義。

1 材料和方法

1.1 材料

供試40批紅花樣品均為菊科植物紅花的干燥管狀花,于花由黃變紅時(shí)采集,產(chǎn)地信息見(jiàn)表1。采集的紅花樣品在產(chǎn)地陰干后粉碎,過(guò)50目篩,干燥保存。憑證標(biāo)本保存于南京中醫(yī)藥大學(xué)江蘇省中藥資源產(chǎn)業(yè)化過(guò)程協(xié)同創(chuàng)新中心。所有紅花樣品經(jīng)南京中醫(yī)藥大學(xué)藥學(xué)院嚴(yán)輝副教授鑒定。

含有Hg、Pb、Cu、As、Cd、Na、Mg、P、K、Cr、Mn、Fe、Ni、Zn、Sr、B、Li、Be、Tl、Bi、Sc、Ti、V、Co、Ga、Y、Ba和Al的多元素標(biāo)準(zhǔn)溶液(濃度均為100 μg·mL-1)(批號(hào)GSB 04-1767-2004)以及內(nèi)標(biāo)溶液73Ge(批號(hào)GSB 04-1728-2004)、115In(批號(hào)GSB04-1731-2004)和209Bi(批號(hào)GSB 04-1719-2004)的單元素標(biāo)準(zhǔn)溶液(濃度均為1 000 μg·mL-1)購(gòu)自國(guó)家有色金屬及電子材料分析測(cè)試中心;硝酸(優(yōu)級(jí)純,批號(hào)180725)和雙氧水(優(yōu)級(jí)純,批號(hào)180819)購(gòu)自上海國(guó)藥化學(xué)試劑公司;屈臣氏蒸餾水。

1.2 方法

1.2.1 樣品采集 采用五點(diǎn)取樣法,分別在田塊(10 m×5 m)2條對(duì)角線的交點(diǎn)以及交點(diǎn)到4個(gè)角的中間點(diǎn)采樣,每個(gè)田塊采摘約2 kg管狀花,混勻后作為一批樣品。每批樣品3個(gè)田塊。

1.2.2 標(biāo)準(zhǔn)溶液制備方法 分別精密吸取100 μg·mL-1多元素標(biāo)準(zhǔn)溶液200 μL,用蒸餾水定容至10 mL,配制成含有Hg、Pb、Cu、As、Cd、Na、Mg、P、K、Cr、Mn、Fe、Ni、Zn、Sr、B、Li、Be、Tl、Bi、Sc、Ti、V、Co、Ga、Y、Ba和Al的混合標(biāo)準(zhǔn)母液,質(zhì)量濃度為2 μg·mL-1,逐級(jí)稀釋配制系列質(zhì)量濃度的標(biāo)準(zhǔn)溶液。

1.2.3 樣品溶液制備方法 精密稱取紅花樣品粉末0.2 g,置于聚四氟乙烯消解罐中,加入硝酸7 mL和雙氧水3 mL,靜置10 min,待反應(yīng)不劇烈,加蓋密封,放入Milestone Ethos D型微波消解儀(意大利Milestone公司)中,消解程序?yàn)椋航?jīng)10 min由室溫升溫至180 ℃;經(jīng)5 min由180 ℃升溫至220 ℃,于220 ℃消解20 min;冷卻至室溫。取出消解罐,在通風(fēng)櫥中將酸揮干,用蒸餾水定容至50 mL作為樣品溶液。以硝酸7 mL和雙氧水3 mL混合后作為空白。

1.2.4 樣品測(cè)定方法 樣品溶液注入OptimaTM2100 DV電感耦合等離子體發(fā)射光譜儀(美國(guó)Perkin Elmer公司)進(jìn)行測(cè)定,測(cè)定條件:等離子體射頻功率1.3 kW;冷卻氣流速15 L·min-1,載氣流速0.8 L·min-1,輔助氣流速0.2 L·min-1;觀測(cè)方向Axial;觀測(cè)高度15 nm;1次讀數(shù)時(shí)間5 s;進(jìn)樣延時(shí)30 s;泵速15 r·min-1。每個(gè)樣品重復(fù)測(cè)定3次,結(jié)果取平均值。

借鑒鐵橋峽谷的經(jīng)驗(yàn),位于南京市區(qū)的小型近現(xiàn)代廠礦可采取主題博物館模式,特別是廠區(qū)整體價(jià)值不大,僅個(gè)別建筑具有歷史價(jià)值、科學(xué)價(jià)值、藝術(shù)價(jià)值時(shí),將建筑物改造為主題博物館,可以最大程度地保護(hù)和展示重要信息,同時(shí)不影響廠區(qū)其他區(qū)域的開(kāi)發(fā)。

1.2.5 方法學(xué)考察

1.2.5.1 精密度試驗(yàn) 取質(zhì)量濃度2 μg·mL-1混合標(biāo)準(zhǔn)母液,按上述方法連續(xù)重復(fù)測(cè)定6次。

1.2.5.2 重復(fù)性試驗(yàn) 平行制備6份樣品溶液,按上述方法進(jìn)行測(cè)定。

1.2.5.3 穩(wěn)定性試驗(yàn) 取樣品溶液,分別于制備后0、1、2、4、6、8和12 h按上述方法進(jìn)行測(cè)定。

1.2.5.4 加樣回收率試驗(yàn) 精密稱取同一樣品適量,分別加入已知樣品中各元素含量80%、100%和120%的標(biāo)準(zhǔn)溶液,按上述方法進(jìn)行測(cè)定。

1.3 數(shù)據(jù)處理

以標(biāo)準(zhǔn)溶液的質(zhì)量濃度為橫坐標(biāo)(x)、各標(biāo)準(zhǔn)品峰面積為縱坐標(biāo)(y)計(jì)算各無(wú)機(jī)元素的回歸方程、相關(guān)系數(shù)、線性范圍、檢測(cè)限和定量限。利用SIMCA-P 14.1軟件對(duì)實(shí)驗(yàn)數(shù)據(jù)進(jìn)行聚類分析和主成分分析,并繪制主成分分析散點(diǎn)圖。

2 結(jié)果和分析

2.1 線性關(guān)系考察

28種無(wú)機(jī)元素標(biāo)準(zhǔn)曲線的回歸方程、相關(guān)系數(shù)、線性范圍、檢測(cè)限和定量限見(jiàn)表2。由表2可見(jiàn):28種無(wú)機(jī)元素標(biāo)準(zhǔn)曲線的線性關(guān)系良好,檢測(cè)限和定量限較低。

2.2 方法學(xué)考察

精密度試驗(yàn)中各無(wú)機(jī)元素含量的RSD值為0.49%~4.80%,表明儀器精密度良好。重復(fù)性試驗(yàn)中各無(wú)機(jī)元素含量的RSD值為1.06%~4.50%,表明該方法重復(fù)性良好。穩(wěn)定性試驗(yàn)中各無(wú)機(jī)元素含量的RSD值小于4.98%,表明樣品溶液在12 h內(nèi)穩(wěn)定。加樣回收率試驗(yàn)中加樣回收率為96.80%~105.70%,RSD值小于4.00%,表明該方法準(zhǔn)確可靠。

2.3 供試40批紅花樣品中無(wú)機(jī)元素含量分析

供試40批紅花樣品中重金屬元素、大量元素、必需微量元素和其他12種無(wú)機(jī)元素的含量分別見(jiàn)表3、表4、表5和表6。

由表3可見(jiàn):供試40批紅花樣品的重金屬元素中Cu含量均最高;Hg和Pb含量也較高;As在來(lái)自甘肅瓜州市的S4和S5樣品,來(lái)自甘肅玉門市的S7、S8、S10、S11、S12、S13和S14樣品,來(lái)自新疆裕民縣的S25樣品以及來(lái)自云南永勝縣的S38樣品中檢出;Cd僅在來(lái)自云南永勝縣的S40樣品中檢出。

由表4可見(jiàn):供試40批紅花樣品的大量元素中K含量(19.961~62.413 mg·g-1)最高,P含量(2.479~4.898 mg·g-1)和Mg含量(1.283~3.074 mg·g-1)較低,Na含量(0.155~1.212 mg·g-1)最低。

由表5可見(jiàn):供試40批紅花樣品的必需微量元素中Fe含量(196.443~1 407.164 μg·g-1)和B含量(55.552~136.285 μg·g-1)較高,Ni含量(0.181~2.635 μg·g-1)最低。

由表6可見(jiàn):供試40批紅花樣品中,Al含量(149.168~1 235.769 μg·g-1)較高,來(lái)自江蘇和安徽的紅花樣品均未檢出Sc、Co和Y,來(lái)自新疆和云南的部分紅花樣品也未檢出Sc、Co和Y。

2.4 不同省(自治區(qū))紅花樣品中無(wú)機(jī)元素含量分析

不同省(自治區(qū))紅花樣品中重金屬元素、大量元素和必需微量元素的含量分別見(jiàn)表7、表8和表9。

由表7可見(jiàn):來(lái)自云南的紅花樣品中Hg含量最高,顯著高于其他省(自治區(qū))的紅花樣品,而來(lái)自江蘇的紅花樣品中Hg含量最低;來(lái)自云南和甘肅的紅花樣品中Pb含量較高,顯著高于來(lái)自江蘇的紅花樣品;來(lái)自不同省(自治區(qū))的紅花樣品中Cu含量無(wú)顯著差異;來(lái)自不同省(自治區(qū))的紅花樣品中As和Cd含量較低或未檢出。

由表8可見(jiàn):來(lái)自甘肅的紅花樣品中Na和Mg含量顯著高于其他省(自治區(qū))的紅花樣品;來(lái)自云南和新疆的紅花樣品中P含量顯著高于其他省(自治區(qū))的紅花樣品;來(lái)自江蘇的紅花樣品中K含量最高,來(lái)自云南的紅花樣品中K含量顯著低于其他省(自治區(qū))的紅花樣品。

由圖1可見(jiàn):總體上看,來(lái)自同一省(自治區(qū))的紅花樣品能夠較好地聚為一類,其中,來(lái)自安徽和江蘇的紅花樣品先聚在一起,來(lái)自甘肅和新疆的紅花樣品距離較近,來(lái)自云南的紅花樣品與其他省(自治區(qū))的紅花樣品距離較遠(yuǎn)。

2.6 供試40批紅花樣品的主成分分析

主成分分析結(jié)果顯示:前4個(gè)主成分的累計(jì)方差貢獻(xiàn)率達(dá)到81.4%,因此,提取前4個(gè)主成分繪制主成分分析散點(diǎn)圖,結(jié)果見(jiàn)圖2。由圖2可見(jiàn):總體上看,除來(lái)自云南的紅花樣品分布較分散外,來(lái)自同一省(自治區(qū))的紅花樣品能夠較好地聚集,與聚類分析結(jié)果一致。

3 討論和結(jié)論

人體吸收利用無(wú)機(jī)元素有2條途徑,一是隨膳食直接攝入而吸收,二是使用溶劑提取后再被添加利用[16]。紅花作為藥食兩用植物,所含的無(wú)機(jī)元素與其營(yíng)養(yǎng)價(jià)值和安全性息息相關(guān),目前,紅花已被國(guó)家衛(wèi)生健康委員會(huì)列入可作為保健食品的中藥名單,開(kāi)發(fā)前景廣闊。根據(jù)WM 2—2001《藥用植物及制劑進(jìn)出口綠色行業(yè)標(biāo)準(zhǔn)》,綠色藥用植物中重金屬總含量應(yīng)小于等于20.0 mg·kg-1,Cu含量小于等于20.0 mg·kg-1,Hg含量小于等于0.2 mg·kg-1,Pb含量小于等于5.0 mg·kg-1,Cd含量小于等于0.3 mg·kg-1,As含量小于等于2.0 mg·kg-1,40批紅花樣品中Hg含量均超標(biāo),Pb、As和Cd含量均不超標(biāo),來(lái)自安徽碭山縣的S1樣品中Cu含量超標(biāo)。舒鳳等[17]研究發(fā)現(xiàn),在無(wú)任何消除Hg記憶效應(yīng)的措施下,0.5 μg·L-1Hg即可產(chǎn)生明顯的記憶效應(yīng),且隨著Hg濃度的增加,記憶效應(yīng)不斷增強(qiáng)。本研究中,供試40批紅花樣品的重金屬元素中Hg含量均超標(biāo),推測(cè)主要由于實(shí)驗(yàn)過(guò)程中未采取消除Hg記憶效應(yīng)的措施。來(lái)自不同省(自治區(qū))的紅花樣品中,來(lái)自云南和甘肅的紅花樣品中Pb含量較高,除顯著高于來(lái)自江蘇的紅花樣品外,各省(自治區(qū))的紅花樣品中Pb含量總體上差異不顯著,可能是由于紅花對(duì)Pb的吸收與環(huán)境中Pb濃度無(wú)關(guān)[18]。供試紅花樣品所含的重金屬元素中Cu含量最高,與田大年等[19]的研究結(jié)果一致,可能是因?yàn)橹仓暝谏L(zhǎng)過(guò)程中需要從土壤中吸收Cu離子,以保證正常的光合作用及生長(zhǎng)[20-21]。

供試紅花樣品所含的大量元素中K含量最高,P和Mg的含量居中,這可能與紅花選擇性吸收這3種大量元素有關(guān)。賈宏濤等[22]研究認(rèn)為,紅花是一種需P量中等、需K量較多的植物,且在生長(zhǎng)過(guò)程中,對(duì)K的吸收以苗期和花蕾期較大,尤其是花對(duì)K的需求量最大[13,23],這可能是紅花樣品中K和P含量高于其他大量元素的原因。

在聚類分析圖中,聚集為一類的樣品中無(wú)機(jī)元素的組成相似。聚類分析和主成分分析結(jié)果表明:來(lái)自甘肅的紅花樣品能較好地聚集在一起,推測(cè)由于來(lái)自甘肅的紅花樣品大多來(lái)自于基地,且種植品種較為單一。來(lái)自新疆的紅花樣品相對(duì)較離散,可能與新疆種植的紅花品種較多(300多個(gè)品種)[8]有關(guān),此外,新疆的地域廣闊和種植方式多樣也是導(dǎo)致樣品離散的潛在原因。新疆屬于低降水量的溫帶大陸性氣候,優(yōu)質(zhì)的紅花種植區(qū)主要集中在海拔1 000~1 250 m[8],而云南屬熱帶季風(fēng)氣候,紅花主要種植區(qū)(如永勝縣)的平均海拔在1 300 m左右,氣候良好,降水穩(wěn)定[24],不同的氣候和生態(tài)條件導(dǎo)致云南的紅花樣品中無(wú)機(jī)元素組成與其他地區(qū)產(chǎn)生了較大的差異。

本研究采用微波消解法和ICP-AES法分析不同產(chǎn)地40批紅花樣品中28種無(wú)機(jī)元素,建立了一種快速、準(zhǔn)確分析紅花中無(wú)機(jī)元素的方法。本研究結(jié)果顯示:紅花富含多種對(duì)人體有益的大量元素K、P和Na以及微量元素Fe、B和Zn。但本研究未對(duì)有益元素Ca和Se進(jìn)行檢測(cè),后期實(shí)驗(yàn)需對(duì)其進(jìn)行檢測(cè)。此外,還需要加強(qiáng)監(jiān)測(cè)紅花中重金屬元素含量,對(duì)土壤的重金屬污染進(jìn)行治理。不同產(chǎn)地的紅花樣品中無(wú)機(jī)元素含量存在一定差異,因此,需要針對(duì)不同產(chǎn)地,在紅花的生長(zhǎng)過(guò)程中使用不同類型的肥料,從而縮小產(chǎn)地間的差異,提高市場(chǎng)上紅花藥材的整體質(zhì)量。另外,土壤中無(wú)機(jī)元素含量與紅花中無(wú)機(jī)元素含量間的關(guān)系也需要進(jìn)一步探究。
