人類及小鼠某些細(xì)胞系培養(yǎng)中苯基桿菌的污染

目的:對細(xì)胞培養(yǎng)體系中出現(xiàn)的桿狀污染物進行分離和鑒定,并探討如何清除該污染物.方法:采用固體培養(yǎng)基平板劃線法分離細(xì)菌株,通過熒光染色和透射電鏡對其進行形態(tài)學(xué)觀察;結(jié)合16S rRNA基因序列分析,進行菌株鑒定;用生長狀態(tài)良好的細(xì)胞上清復(fù)蘇凍存的已經(jīng)污染的細(xì)胞,檢測細(xì)胞復(fù)蘇的存活率.結(jié)果:該污染物經(jīng)形態(tài)學(xué)和16S rRNA基因序列鑒定為苯基桿菌.形態(tài)學(xué)觀察表明它有一個二態(tài)生命周期:即游動期和附著期.大多數(shù)情況下該菌可以與宿主細(xì)胞共生,常規(guī)抗生素均不能徹底清除該細(xì)菌.采用生長狀態(tài)良好的細(xì)胞上清復(fù)蘇凍存細(xì)胞可以明顯提高了細(xì)胞的存活率.結(jié)論:本實驗報導(dǎo)了苯基桿菌的二態(tài)生命周期,同時我們發(fā)現(xiàn)用細(xì)胞上清復(fù)蘇凍存細(xì)胞可以顯著提高細(xì)胞的存活率.
