金線吊烏龜Stephania cepharanthaHayata 為防己科千金藤屬植物,主要分布于我國西南、東南等地。民間常以其塊根入藥,又名白藥子、白大藥、山烏龜?shù)?,味苦、性寒,具有清熱解毒、散瘀止痛等功效,常用于治療咽喉腫痛、風(fēng)濕痹痛、腮腺炎等疾?。?-4]。該植物塊根的甲醇提取物、氯仿可溶的生物堿部分具有抗病毒作用;已從塊根、莖葉和種子中分離得到多種類型的生物堿,具有抗炎、抗癌細(xì)胞生長、抗骨肉瘤等多種臨床藥理活性[3-13]。為進(jìn)一步研究金線吊烏龜?shù)幕瘜W(xué)成分,本實(shí)驗(yàn)對(duì)其塊根部分進(jìn)行了提取與分離,得到15個(gè)化合物,其中化合物13 為新天然產(chǎn)物,化合物8~9、13 為首次從該植物中分離得到。

金線吊烏龜藥材購于浙江寧波德康生物制品有限公司,產(chǎn)地為四川省,經(jīng)復(fù)旦大學(xué)藥學(xué)院康云講師鑒定為防己科千金藤屬植物金線吊烏龜Stephania cepharanthaHayata 的干燥塊根,標(biāo)本保存于上海市復(fù)旦大學(xué)藥學(xué)院。
金線吊烏龜塊根粗粉6 kg,室溫下以95%乙醇浸泡、滲漉提取,減壓回收溶劑,得浸膏約800 g。浸膏以2%鹽酸水溶液分散均勻,用石油醚脫脂,以氨水調(diào)節(jié)到pH =10,用二氯甲烷萃取3 次,減壓濃縮萃取液,得二氯甲烷萃取物200 g,水層再用正丁醇萃取,減壓濃縮得正丁醇萃取物24 g。二氯甲烷萃取物經(jīng)正相硅膠柱,以二氯甲烷-甲醇溶劑系統(tǒng)(80∶1、50∶1、25∶1、14∶1、1∶1、0∶1) 梯度洗脫,再經(jīng)反復(fù)RP-C18柱層析(甲醇-水)、制備型高效液相色譜(RP-C18色譜柱,甲醇-水或乙腈-水) 以及制備TLC 等方法分離,得到化合物1(15 mg)、2(5 mg)、3(380 mg)、4(150 mg)、5(5 mg)、6(4.5 mg)、7(3 mg)。正丁醇部位萃取物進(jìn)行大孔樹脂HZ818 柱層析,用乙醇-水(3∶7、5∶5、7∶3、95∶5) 梯度洗脫,再經(jīng)反復(fù)Sephadex LH-20 柱層析(甲醇-水)、RP-C18開放柱層析(甲醇-水) 以及制備型高效液相色譜(RP-C18色譜柱,甲醇-0.02%二乙胺-水溶液)等方法分離,得到化合物8(2 mg)、9(6 mg)、10(27 mg)、11(4 mg)、12(5 mg)、13(8 mg)、14(7 mg)、15(1.1 mg)。

不同產(chǎn)地植物的次生代謝產(chǎn)物常有差異,本實(shí)驗(yàn)選擇鮮有報(bào)道的四川產(chǎn)金線吊烏龜進(jìn)行研究,以期豐富該種類的化學(xué)成分。金線吊烏龜?shù)亩喾N生物堿具有抗腫瘤、抗炎等作用[7-13],本研究所得化合物可用于相關(guān)活性的篩選,以期有助于研究類似結(jié)構(gòu)生物堿的構(gòu)效關(guān)系,進(jìn)一步闡明該植物的藥效物質(zhì)基礎(chǔ)。
