摘要 [目的] 合成并鑒定氟苯尼考胺人工抗原。[方法] 采用水解的方法制備了氟苯尼考胺,并通過紫外、紅外、質(zhì)譜鑒定氟苯尼考胺水解成功。采用戊二醛法合成了氟苯尼考人工抗原,并通過紫外光譜進(jìn)行鑒定。免疫大白兔制備了氟苯尼考胺高特異性抗體。[結(jié)果] 通過紫外、紅外、質(zhì)譜鑒定氟苯尼考胺水解成功,抗體效價達(dá)6.4×104。[結(jié)論] 氟苯尼考胺人工抗原合成成功,為建立FFA免疫化學(xué)方法奠定了技術(shù)基礎(chǔ)。

Abstract [Objective] The research aimed to prepare and identify the artificial antigen of florfenicol amine. [Method] The artificial antigen of florfenicol amine was synthesized by using hydrolysis method. The product was examined by ultraviolet spectrum, infrared spectrum and mass. FFA was covalently attached to carrier protein as immunogen by glutaraldehyde method. The conjugates was identified with ultraviolet spectrum. FFABSA was used to immune New Zealand white rabbits and prepare polyclonal antibody of FFA with high affinity and high specificity.[Result] The results showed these products were synthesized successfully. The antibodies produced with FFABSA conjugate showed titer of 6.4×104. [Conclusion] The synthesis route of FFA artificial antigen was feasible. This research could lay the foundation for further establishment of the immune detection method for FFA.

氟苯尼考(Florfenicol,結(jié)構(gòu)式見圖1)是替代禁用藥物氯霉素的一種理想的藥物,現(xiàn)已廣泛應(yīng)用于水產(chǎn)養(yǎng)殖的防病抗病上[1-3]。氟苯尼考在動物組織中的主要代謝產(chǎn)物為氟苯尼考胺(florfenicol amine,F(xiàn)FA,結(jié)構(gòu)式見圖1),許多國家以氟苯尼考胺作為使用氟苯尼考的殘留標(biāo)示物[4]。盡管氟苯尼考毒性低于氯霉素,但由于不正確或超量的使用,勢必造成氟苯尼考在畜禽水產(chǎn)品中的殘留,特別是現(xiàn)已出現(xiàn)了對氟苯尼考耐藥的菌株,而且氟苯尼考的用量呈上升的趨勢,這對人類的身體健康造成了潛在的威脅。因此,研究其殘留監(jiān)控技術(shù)對保障我國人民身體健康和水產(chǎn)品食用安全具有重要意義。目前國內(nèi)外對氟苯尼考的檢測主要有HPLC、LCMS
等方法[5-10],儀器方法雖然靈敏度高、準(zhǔn)確性好,但需要昂
貴的儀器,檢測成本較高,不適合大量樣品的篩選。酶聯(lián)免疫吸附測定法(ELISA)因具有快速、靈敏、高效等優(yōu)點,用于大批量藥物殘留篩選越來越廣泛。國內(nèi)外生物公司已經(jīng)開始研發(fā)氟苯尼考檢測試劑盒,但這些試劑盒檢測的靶物質(zhì)為氟苯尼考原型藥,不能完成其代謝產(chǎn)物氟苯尼考胺的檢測[11]。到目前為止我國沒有自行合成的氟苯尼考胺標(biāo)準(zhǔn)品,這制約了我國研究氟苯尼考胺,也未見有關(guān)氟苯尼考胺ELISA試劑盒面市。筆者擬采用化學(xué)方法水解氟苯尼考制備氟苯尼考胺標(biāo)準(zhǔn)品,合成氟苯尼考胺人工抗原,以期為制備氟苯尼考胺的特異性抗體和建立氟苯尼考胺ELISA檢測方法提供技術(shù)支撐。
3 結(jié)論
該研究采用水解的方法,制備了氟苯尼考胺,合成了氟苯尼考胺人工抗原,為制備針對FFA特異性抗體和建立氟苯尼考?xì)埩魳?biāo)示物FFA檢測方法提供了技術(shù)技術(shù)支撐。武漢輕工大學(xué)食品科學(xué)與工程學(xué)院實驗室目前采用最靈敏的特異性抗體(R5)已建立了檢測FFA間接競爭ELISA 方法(成果另文發(fā)表),為我國開發(fā)FFA免疫檢測試劑盒打下堅實的基礎(chǔ),具有一定的創(chuàng)新性和經(jīng)濟(jì)價值。
