Effects of fermented mixed residue replacing part of diet on the diversity index of intestinal microflora of Fufeng

Abstract:[Objective]The objective of this study was to assess the effects of four fermented mixed residues(Siraitia grosvenori residue,tofu residue,sweet potato peel and cocoyam powder)replacing part of diet on intestinal microflora of Fufeng Ma-chickens in Guangxi.At the same time,the optimal ratio of fermented mixed residue replacing part of diet was evaluated.and the reference basis for the application of four fermented mixed residue in chicken breeding was pro-vided.【Method]Twenty 60-day-old Fufeng Ma-chickens in Guangxi(female)were selected and randomly divided into 4 groups with 5 chickens in each group.The diets of 5%(R5),10%(R10),and 15%(R15)were replaced with fermented mixed residue of S.grosvenori residue,tofu residue,sweet potato peel and cocoyam powder respectively.Thewhole diet was used as the control group(CK)in afeeding trial for 97 d.After feeding,the contents of the Fufeng Ma-chickens' large intestine were collected,and effectsof fermented mixed residue replacingpart of diet on the diversity indexof intesti-nal microflora ofFufeng Ma-chickens in Guangxi were studied based on Illumina high-throughputsequencing technology【Result]The Chaol index of intestinal microflora of Fufeng Ma-chickens in Guangxi was ranked as R15gt;CKgt;R10gt;R5,Shannon index was ranked as CKgt;R15gt;R10gt;R5,and Simpson index was ranked as CK=R15=R10gt;R5.There was no significant difference in the Chao 1 index,Shannon index,and Simpson index between each experimental group and CK (Pgt;0.05,the same as below).At the level of phylum classification,among which Bacteroidetes,F(xiàn)irmicutes,and Proteo-bacteria were the dominant bacteria.The relative abundance of Bacteroidetes was ranked as R10gt;R15gt;CKgt;R5,F(xiàn)ir micuteswas ranked as R5gt;CKgt;R15gt;R10,and Proteobacteria was ranked as CKgt;R10gt;R15gt;R5.Among them,therela- tive abundance of Proteobacteria and Epsilonbacteraeota in the R5 was significantly lower than that of CK(Plt;0.05,the same as below).At the level of genus classification,the relative abundance of Proteus was rank as CKgt;R10gt;R5gt;R15.The relative abundance of the threealternative test groups was significantly lower than that of CK.The relative abundance of Ruminiclostridium 9 was ranked as R10gt;R15gt;CKgt;R5,and only the relative abundance of R10 group was signifi-cantly higher than that of the CK.The relative abundance of Faecalibacterium was ranked as CKgt;R10gt;R5gt;R15,and the relative abundance of R15 group was significantly lower than that of the CK.【Conclusion]Replacing 5%,10%and 15%diet by four fermented mixed residue of S.grosvenori,tofu,sweet potato peel and cocoyam powder for Fufeng Ma-chickens in Guangxi do not affect the species abundance and diversity of intestinal microflora of chickens.Based on the comprehensive evaluation of beneficial bacteria and harmful bacteria,10%addition rate can increase the abundance of probioticRumimiclostridium_9 and has the least effect on Protens,which is the optimal addition rate.

【研究意義】廣西屬于亞熱帶氣候,農(nóng)林物產(chǎn)豐富,但每年產(chǎn)出的大量農(nóng)林副產(chǎn)品,如香蕉莖葉渣、甘蔗渣、木薯渣及紅薯藤等均未得到充分利用(李茂等,2015)。羅漢果渣、豆腐渣、紅薯皮和芋頭粉等加工副產(chǎn)品營(yíng)養(yǎng)豐富,具有作為飼料資源開發(fā)的潛力(歐榮娣,2015;王啟芝等,2018;劉立軍等,2020;梁麗芬等,2022),若能有效利用這些加工副產(chǎn)品將極大豐富廣西的飼料資源。腸道微生物對(duì)畜禽的生產(chǎn)性能、健康狀況及營(yíng)養(yǎng)消化吸收等具有重要意義(辛可啟等,2020;顧艷麗等,2022),日糧組成、基因型及養(yǎng)殖環(huán)境等則影響腸道微生物的群落結(jié)構(gòu)和功能特性(周俊華等,2021)。因此,研究日糧組成、養(yǎng)殖環(huán)境和飼養(yǎng)方式等因素對(duì)畜禽腸道微生物的影響,對(duì)進(jìn)一步調(diào)控腸道微生物及提高畜禽生產(chǎn)性能具有重要意義。【前人研究進(jìn)展】腸道微生物群落結(jié)構(gòu)高度依賴于作為最終代謝底物來源的日糧(Rehman et al.,2007)。目前,有關(guān)日糧中添加不同農(nóng)林副產(chǎn)品對(duì)雞腸道微生物影響的研究已較多報(bào)道。張耀(2007)研究報(bào)道,在飼料中添加陳皮可顯著促進(jìn)白羽肉雞盲腸乳酸桿菌的生長(zhǎng),并提高回盲腸中乳酸桿菌和大腸桿菌的相對(duì)豐度;馮玉升和魏時(shí)來(2015)發(fā)現(xiàn)在飼糧中添加8%和16%啤酒花渣能顯著降低腸道乳酸菌數(shù)量,但對(duì)腸道大腸桿菌數(shù)量的影響不明顯;鄺哲師等(2016)研究表明,桑葉粉具有降低胡須雞盲腸乳酸菌數(shù)量的趨勢(shì),而發(fā)酵桑葉粉有提高乳酸菌數(shù)量的趨勢(shì),但與對(duì)照組相比其差異均不顯著;王自豪等(2022)用發(fā)酵桂閔引象草替代部分日糧,發(fā)現(xiàn)雞腸道菌群的物種豐度增高、菌群多樣性降低,主要是由于變形菌門和互養(yǎng)菌門的相對(duì)豐度升高;張莉等(2022)研究發(fā)現(xiàn),飼糧中添加藍(lán)藻粉會(huì)改變崇仁麻雞腸道菌群物種多樣性,適量(≤10%)的添加有利于改善盲腸菌群結(jié)構(gòu),具體表現(xiàn)為:提高厚壁菌門細(xì)菌數(shù)量,降低擬桿菌門/厚壁菌門細(xì)菌比例,增加腸道內(nèi)乳桿菌等有益微生物的數(shù)量,同時(shí)降低脫硫弧菌等有害微生物的數(shù)量。此外,在日糧中添加青貯柑橘渣(易蘭蘭等,2021)、艾草粉(歸榮等,2022)、黃粉蟲粉(蔣騰等,2022)、大麥蟲粉(余親平等,2022)、羅漢果渣(梁麗芬,2022;滕少花等,2023)、辣木葉粉(雷福紅等,2023)等其他農(nóng)林副產(chǎn)品對(duì)雞腸道微生物群落均有一定影響,并證實(shí)不同日糧組成有些能改變某種特定菌群的相對(duì)豐度,但宏觀上不影響雞腸道微生物群落多樣性;有些則很大程度上影響某些特定菌群的相對(duì)豐度或改變物種數(shù)量,從而影響雞腸道微生物群落多樣性。【本研究切入點(diǎn)】雞腸道微生物群落多樣性因日糧組成不同而存在差異,但至今鮮見以羅漢果渣、豆腐渣、紅薯皮和芋頭粉的單一組分或4種組分混合作為非糧飼料來源替代部分日糧對(duì)雞腸道微生物群落影響的研究報(bào)道?!緮M解決的關(guān)鍵問題】以廣西富鳳麻雞大腸內(nèi)容物菌群DNA為研究對(duì)象,利用高通量測(cè)序技術(shù)對(duì)16S rDNA序列V3~V4可變區(qū)進(jìn)行測(cè)序,分析大腸菌群的物種組成及相對(duì)豐度差異,明確羅漢果渣、豆腐渣、紅薯皮和芋頭粉4種組分混合渣發(fā)酵料替代部分日糧對(duì)雞腸道微生物菌群的影響,同時(shí)評(píng)估混合渣發(fā)酵料替代日糧的最佳比例,為混合渣發(fā)酵料在雞養(yǎng)殖生產(chǎn)中的應(yīng)用提供參考依據(jù)。
1材料與方法
1.1混合渣發(fā)酵料制備
混合渣成分為羅漢果渣、豆腐渣、紅薯皮和芋頭粉,其配比為7.0:1.0:1.5:0.5?;旌显砑幽蠈幬⑷鹕锟萍加邢薰旧a(chǎn)的EM菌青貯飼料專用發(fā)酵劑,經(jīng)厭氧發(fā)酵45d后使用。
1.2試驗(yàn)設(shè)計(jì)
在廣西畜牧研究所實(shí)驗(yàn)雞場(chǎng)挑選遺傳背景相近、體重相近、體況良好的60日齡自繁廣西富鳳麻雞(母雞)20羽,隨機(jī)分成4組,每組5羽。其中,3個(gè)試驗(yàn)組以混合渣發(fā)酵料替代部分日糧,替代比例分別為5%(R5)、10%(R10)和15%(R15),以全日糧試驗(yàn)組為對(duì)照(CK)。試驗(yàn)周期97 d,其中預(yù)飼期7 d、正式期90d。日糧原料組成及營(yíng)養(yǎng)水平參照滕少花等(2023)的研究方法。試驗(yàn)雞群自由飲水、自由采食,其他按雞場(chǎng)日常生產(chǎn)要求進(jìn)行管理。
1.3樣品采集及DNA提取
將各處理組廣西富鳳麻雞靜脈放血處死,以5%新潔爾滅消毒液全身消毒(浸泡5 min)后,無菌操作剖開腹腔,玻璃棒刮取大腸壁內(nèi)容物,并迅速放入滅菌離心管中,液氮速凍,-80℃冰箱保存。采用十六烷基三甲基溴化銨(CTAB)法對(duì)各大腸內(nèi)容物樣本進(jìn)行菌群DNA提取,以1%瓊脂糖凝膠電泳檢測(cè)DNA提取質(zhì)量,并通過Qubit(Invitrogen,美國(guó))對(duì)DNA進(jìn)行定量分析。
1.416S rDNA擴(kuò)增測(cè)序及多樣性分析
根據(jù)細(xì)菌16SrDNA序列V3~V4可變區(qū)設(shè)計(jì)引物,正向引物341F:5'-CCTACGGGNGGCWGCAG-3',反向引物805R:5'-GACTACHVGGGTATCTAATCC-3',并在引物上添加測(cè)序通用接頭和樣本特異性Barcode序列。擴(kuò)增程序:98℃預(yù)變性30 s;98℃10 s,54℃30 s,72℃45s,進(jìn)行35個(gè)循環(huán);72℃延伸10 min。PCR擴(kuò)增產(chǎn)物經(jīng)2%瓊脂糖凝膠電泳檢測(cè)后,采用AMPure XT beads回收試劑盒對(duì)目標(biāo)片段進(jìn)行純化回收,然后使用Agilent 2100生物分析儀和Illumina文庫定量試劑盒進(jìn)行評(píng)估,合格的文庫濃度須在2 nmol/L以上。合格的測(cè)序文庫(Index序列不可重復(fù))進(jìn)行梯度稀釋,以NaOH處理變性為單鏈后,以NovaSeq 6000測(cè)序儀進(jìn)行2×250 bp的雙端測(cè)序,相應(yīng)試劑為NovaSeq 6000 SP Reagent Kit(500 Cycles)。上機(jī)測(cè)序委托杭州聯(lián)川生物技術(shù)股份有限公司完成。
通過對(duì)測(cè)序獲得的Reads進(jìn)行拼接、過濾及操作分類單元(OTU)聚類,并基于SILVA(Release 132)和NT-16S數(shù)據(jù)庫進(jìn)行序列比對(duì),以獲得分類學(xué)上的物種注釋。根據(jù)物種注釋結(jié)果,選取每組樣本在門和屬分類水平上相對(duì)豐度排名靠前的物種進(jìn)行比較分析;對(duì)OTUs進(jìn)行Alpha多樣性分析,比較樣品間的差異并進(jìn)行統(tǒng)計(jì)學(xué)檢驗(yàn),其中兩組間差異分析采用Wilcoxon軼和檢驗(yàn)。
2結(jié)果與分析
2.1廣西富鳳麻雞大腸微生物群落OTUs分析結(jié)果
對(duì)測(cè)序獲得的原始數(shù)據(jù)進(jìn)行雙端拼接、質(zhì)量控制及嵌合體過濾,結(jié)果從CK、R5、R10、R15試驗(yàn)組菌群總DNA中分別獲得68259、68299、69173和71765條有效序列。采用Vsearch 2.21.1按照97%的序列相似度將有效序列聚類為OTUs(Blaxter et al.,2005),結(jié)果(圖1)表明,CK、R5、R10、R15試驗(yàn)組分別獲得2774、2886、3967和3686個(gè)OTUs,對(duì)應(yīng)的特有OTUs分別為1534、1606、2527和2300個(gè),共有OTUs為619個(gè)。
由圖2可看出,CK、R5、R10和R15試驗(yàn)組廣西富鳳麻雞大腸微生物群落OTUs稀釋曲線均隨著測(cè)序數(shù)據(jù)量的增大而趨于平緩,表明樣本高通量測(cè)序數(shù)據(jù)量合理,測(cè)序數(shù)據(jù)量足夠反映樣品中的物種組成特征,可用于后續(xù)研究。
2.2廣西富鳳麻雞大腸微生物群落Alpha多樣性分析結(jié)果
由表1可知,CK、R5、R10和R15試驗(yàn)組廣西富鳳麻雞大腸微生物群落的覆蓋度均為100%,說明測(cè)序深度能全面反映樣品的菌群多樣性。Chaol指數(shù)表征群落物種豐富度,Shannon指數(shù)和Simpson指數(shù)則反映群落結(jié)構(gòu)多樣性。廣西富鳳麻雞大腸微生物群落的Chaol指數(shù)排序?yàn)镽15gt;CKgt;R10gt;R5,Shan-non指數(shù)排序?yàn)镃Kgt;R15gt;R10gt;R5,Simpson指數(shù)排序?yàn)镃K=R15=R10gt;R5。各試驗(yàn)組間的Chaol指數(shù)、Shannon指數(shù)和Simpson指數(shù)均無顯著差異(Pgt;0.05,下同),說明混合渣發(fā)酵料替代5%、10%和15%日糧對(duì)廣西富鳳麻雞大腸微生物群落的物種豐度及多樣性并無顯著影響。
2.3混合渣發(fā)酵料代替部分日糧對(duì)廣西富鳳麻雞大腸微生物門分類水平的影響
如表2所示,廣西富鳳麻雞大腸微生物群落的所有OTUs共注釋到27個(gè)菌門。其中,擬桿菌門(Bac-teroidetes)、厚壁菌門(Firmicutes)、變形菌門(Proteo-bacteria)的相對(duì)豐度之和在CK、R5、R10、R15試驗(yàn)組廣西富鳳麻雞大腸微生物群落中的占比分別為82.87%、84.05%、80.97%和77.55%,屬于絕對(duì)優(yōu)勢(shì)菌門。對(duì)相對(duì)豐度gt;1.00%的菌門進(jìn)行組間差異分析并經(jīng)Wilcoxon檢驗(yàn),結(jié)果顯示變形菌門和Epsilon-bacteraeota的組間差異達(dá)顯著水平(Plt;0.05,下同)。變形菌門相對(duì)豐度排序?yàn)镃Kgt;R 10gt;R 15gt;R5,Epsi-lonbacteraeota相對(duì)豐度排序?yàn)镽10gt;R15gt;CKgt;R5。其中,R5試驗(yàn)組變形菌門和Epsilonbacteraeota的相對(duì)豐度均顯著低于CK。綜上所述,以混合渣發(fā)酵料替代5%日糧可引起廣西富鳳麻雞大腸微生物群落相對(duì)豐度改變,且相對(duì)豐度改變的主要菌門是變形菌門和Epsilonbacteraeota。
2.4混合渣發(fā)酵料替代部分日糧對(duì)廣西富鳳麻雞大腸微生物屬分類水平的影響
廣西富鳳麻雞大腸微生物群落的所有OTUs共注釋到509個(gè)菌屬,相對(duì)豐度前50的菌屬如表3所示。由圖3可知,相對(duì)豐度gt;1.00%的18個(gè)菌屬分別隸屬于4個(gè)菌門。其中,擬桿菌屬(Bacteroides)、理研菌科_RC9_菌群(Rikenellaceae_RC9_gut_group)、擬桿菌目未知菌屬(Bacteroidales_unclassified)、普雷沃氏菌科UCG-001(Prevotellaceae_UCG-001)、Muri-baculaceae_unclassified、擬桿菌門未知菌屬(Bacte-roidetes_unclassified)和普雷沃氏菌科Ga6A1(Pre-votellaceae_Ga6A1_group)隸屬于擬桿菌門,變形桿菌屬(Proteus)和脫硫弧菌屬(Desulfovibrio)隸屬于變形菌門,羅姆布茨菌屬(Romboutsia)、梭菌目未知菌屬(Clostridiales_vadinBB60_group_unclassified)、乳酸桿菌屬(Lactobacillus)、糞桿菌屬(Faecalibacte-rium)、瘤胃菌科UCG-014(Ruminococcaceae_UCG-014)、巨單胞菌屬(Megamonas)、巨球形菌屬(Me-gasphaera)和瘤胃梭菌屬9(Ruminiclostridium_9)隸屬于厚壁菌門;WCHB 1-41_unclassified隸屬于Kiri-timatiellaeota。對(duì)相對(duì)豐度gt;1.00%的菌屬進(jìn)行組間差異分析并經(jīng)Wilcoxon檢驗(yàn),結(jié)果表明變形桿菌屬、瘤胃梭菌屬9及糞桿菌屬存在顯著的組間差異。變形桿菌屬在不同試驗(yàn)組中的相對(duì)豐度排序?yàn)镃Kgt;R10gt;R5gt;R15,瘤胃梭菌屬9在不同試驗(yàn)組中的相對(duì)豐度排序?yàn)镽10gt;R15gt;CKgt;R5,糞桿菌屬在不同試驗(yàn)組中的相對(duì)豐度排序?yàn)镃Kgt;R10gt;R5gt;R15。其中,R5、R10和R15試驗(yàn)組廣西富鳳麻雞大腸微生物群落中的變形桿菌屬相對(duì)豐度均顯著低于CK;R 10試驗(yàn)組的瘤胃梭菌屬9相對(duì)豐度顯著高于CK,而R5和R15試驗(yàn)組的瘤胃梭菌屬9相對(duì)豐度與CK無顯著差異;R15試驗(yàn)組的糞桿菌屬相對(duì)豐度顯著低于CK,而R5和R10試驗(yàn)組的糞桿菌屬相對(duì)豐度與CK無顯著差異。可見,混合渣替代部分日糧引起廣西富鳳麻雞大腸微生物群落相對(duì)豐度改變的主要菌屬是變形桿菌屬、瘤胃松菌屬9和糞桿菌屬。
3討論
雞消化道微生物是由大量革蘭氏陽性菌組成,以擬桿菌門、厚壁菌門和變形菌門為優(yōu)勢(shì)菌門(王晶等,2020)。Wei等(2013)對(duì)公共數(shù)據(jù)庫中3184個(gè)雞(肉仔雞、蛋雞)腸道微生物的16S rDNA序列進(jìn)行分析,結(jié)果發(fā)現(xiàn)厚壁菌門(70.0%)、擬桿菌門(12.3%)、變形菌門(9.3%)的相對(duì)豐度之和在90.0%以上,是雞腸道中的優(yōu)勢(shì)菌門,而梭菌屬、瘤胃球菌屬、乳酸桿菌屬和擬桿菌屬為優(yōu)勢(shì)菌屬。本研究中,擬桿菌門、厚壁菌門、變形菌門在不同試驗(yàn)組的相對(duì)豐度之和為77.55%~84.05%,是廣西富鳳麻雞大腸微生物群落的優(yōu)勢(shì)菌門。動(dòng)物消化系統(tǒng)對(duì)營(yíng)養(yǎng)物質(zhì)的消化和吸收能力部分依賴于腸道微生物群落的物種豐度,而腸道微生物群落的物種豐度決定了腸道代謝過程的范圍和程度(王金全等,2005)。以混合渣發(fā)酵料替代5%、10%、15%日糧不會(huì)影響廣西富鳳麻雞腸道微生物群落的優(yōu)勢(shì)菌門,優(yōu)勢(shì)菌門的穩(wěn)定保證了雞腸道的代謝范圍。在屬分類水平上,擬桿菌屬、理研菌科_RC9_菌群、擬桿菌目未知菌屬、普雷沃氏菌科UCG-001及變形桿菌屬為相對(duì)豐度排名前5的優(yōu)勢(shì)物種,與Wei等(2013)報(bào)道的優(yōu)勢(shì)菌屬存在偏差,可能是腸道菌群對(duì)日糧組成改變的一種響應(yīng),體現(xiàn)了雞腸道代謝過程的程度。但從廣西富鳳麻雞大腸微生物群落Alpha多樣性來看,優(yōu)勢(shì)菌屬的相對(duì)豐度變化不足以引起雞腸道微生物群落物種豐富度及多樣性的變化,說明以混合渣發(fā)酵料替代5%、10%或15%日糧均具有開發(fā)成飼料資源的潛力。本研究中,變形桿菌屬、瘤胃梭菌屬9和糞桿菌屬是混合渣發(fā)酵料替代部分日糧引起廣西富鳳麻雞大腸微生物菌群相對(duì)豐度改變的主要菌屬。其中,變形桿菌屬的相對(duì)豐度排序?yàn)镃Kgt;R10gt;R5gt;R15,瘤胃梭菌屬9的相對(duì)豐度排序?yàn)镽10gt;R15gt;CKgt;R5,糞桿菌屬的相對(duì)豐度排序?yàn)镃Kgt;R10gt;R5gt;R15。這3種菌屬的相對(duì)豐度變化規(guī)律與混合渣發(fā)酵料替代比例的遞增無明顯關(guān)聯(lián)。因此,本研究無法從以上3個(gè)差異菌屬相對(duì)豐度的排序直接評(píng)估篩選出混合渣發(fā)酵料的最優(yōu)添加比例。
腸道益生菌和有害菌對(duì)雞生長(zhǎng)性能、免疫或疾病均產(chǎn)生重要影響。本研究中,益生菌乳酸桿菌屬和有害菌脫硫弧菌屬的相對(duì)豐度在不同試驗(yàn)組中均無顯著差異。瘤胃梭菌屬9、糞桿菌屬和變形桿菌屬是雞腸道中的常見菌屬。甄玉國(guó)等(2020)研究報(bào)道,日糧中添加復(fù)合酵母培養(yǎng)物和乳酸菌培養(yǎng)物可顯著提高肉仔雞盲腸菌群中瘤胃梭菌屬9和糞桿菌屬的相對(duì)豐度;張紅娜等(2020)通過探討解淀粉芽孢桿菌提高蛋雞生產(chǎn)性能的潛在機(jī)制,發(fā)現(xiàn)解淀粉芽孢桿菌可使瘤胃梭菌屬9的相對(duì)豐度顯著增加。日糧中添加益生菌可提高瘤胃梭菌屬9和糞桿菌屬數(shù)量,表明瘤胃梭菌屬9和糞桿菌屬對(duì)雞腸道健康產(chǎn)生積極作用。有關(guān)腸道變形菌門或變形菌屬對(duì)宿主的影響僅在人類中有相關(guān)研究報(bào)道。在人類腸道菌群優(yōu)勢(shì)菌門(厚壁菌門、擬桿菌門、變形菌門和放線菌門)中,以變形菌門最不穩(wěn)定(Faith et al.,2013)。變形菌門對(duì)飲食等環(huán)境因素反應(yīng)敏感(Shin et al.,2015),臨床上通常認(rèn)為腸道中變形菌門的擴(kuò)張可反映腸道微生物群落結(jié)構(gòu)的不穩(wěn)定,且這種不穩(wěn)定結(jié)構(gòu)在非疾病狀態(tài)(新生兒期和胃旁路手術(shù)后)和疾病狀態(tài)(代謝紊亂和腸道炎癥中)下均可觀察到(Morgan et al.,2012;Fei and Zhao,2013;Liou etal.2013;Jakobsson et al.,2014)。目前,有關(guān)變形菌門或變形菌屬對(duì)雞生長(zhǎng)性能、免疫或疾病發(fā)生的影響尚無文獻(xiàn)報(bào)道。本研究中,R10試驗(yàn)組廣西富鳳麻雞大腸中糞桿菌屬、瘤胃梭菌屬9和變形桿菌屬的相對(duì)豐度在3個(gè)混合渣發(fā)酵料替代組中均最高。其中,糞桿菌屬的相對(duì)豐度與CK無顯著差異,瘤胃梭菌屬9的相對(duì)豐度顯著高于CK。從有益菌角度來評(píng)估,以10%的添加比例最優(yōu);從潛在的有害菌角度保守評(píng)估,R10試驗(yàn)組的變形桿菌屬相對(duì)豐度與CK最接近,影響相對(duì)最小。綜上所述,以混合渣發(fā)酵料替代5%、10%和15%日糧不會(huì)影響廣西富鳳麻雞大腸微生物群落的物種豐度及多樣性,其中又以10%的添加比例最優(yōu)。
4結(jié)論
以羅漢果渣、豆腐渣、紅薯皮和芋頭粉4種組分的混合渣發(fā)酵料替代5%、10%和15%日糧飼喂廣西富鳳麻雞,不會(huì)影響雞腸道微生物群落的物種豐度及多樣性;基于有益菌和有害菌的綜合評(píng)估,10%添加比例能增加益生菌瘤胃梭菌屬9相對(duì)豐度且對(duì)變形桿菌屬的影響最小,為最優(yōu)添加比例。
