摘要:合成了甲基對(duì)硫磷的半抗原,并與載體蛋白偶聯(lián)合成了人工抗原,然后通過(guò)免疫的方法制備了其單克隆抗體。在篩選甲基對(duì)硫磷單克隆抗體的基礎(chǔ)上,建立酶聯(lián)免疫檢測(cè)方法,并研制出檢測(cè)蘋(píng)果、玉米、蔬菜中甲基對(duì)硫磷殘留的試劑盒。該試劑盒的靈敏度為0.5 μg/L,對(duì)蘋(píng)果、玉米、蔬菜樣品的最低檢測(cè)限分別可達(dá)到10、20、50 μg/kg;加標(biāo)回收率為70.3%~102.4%,變異系數(shù)為7.4%~13.0%;對(duì)實(shí)際樣品的檢測(cè)結(jié)果與氣相色譜法基本一致。該方法快速、靈敏、可靠,為水果、蔬菜、谷物中甲基對(duì)硫磷的殘留檢測(cè)提供了便捷、準(zhǔn)確的分析手段。

Development of an Enzyme-linked Immunosorbent Assay Kit for Detecting of Parathion-methyl Residues

Abstract:A hapten artificial antigen was prepared, and monoclonal antibodies against parathion-methyl were produced. On the basis of parathion-methyl monoclonal antibody, an enzyme-linked immunosorbent assay(ELISA) kit for parathion-methyl residues in apple, corn and vegetable was developed. The sensitivity of the ELISA kit was 0.5 μg/L. The detection limits for apple, corn, vegetable were 10, 20, 50 μg/kg, respectively. Sample recoveries were 70.3%~102.4%, and the coefficients of variation were 7.4%~13.0%. Results of the ELISA kit were in accordance with those by gas chromatography(GC). This method is rapid, sensitive and reliable, which provides a convenient and accurate way for detection of parathion-methyl residues in fruit, cereal and vegetable.
