摘要 [目的]研究天胡荽的抗腫瘤活性成分。[方法]利用色譜手段進(jìn)行分離純化,應(yīng)用NMR方法鑒定結(jié)構(gòu),并進(jìn)行MTT法抗腫瘤試驗(yàn)。[結(jié)果]從天胡荽中分離鑒定了4種化合物(化合物1~4)。天胡荽的各萃取部位對(duì)LA795細(xì)胞株有一定的抑制作用,其中以正丁醇層萃取物在高濃度800 μg/ml時(shí)的抑制作用較強(qiáng),而石油醚和乙酸乙酯萃取物的抑制作用相對(duì)較弱。單體化合物對(duì)LA795細(xì)胞株的抑制作用均較弱。[結(jié)論]4個(gè)化合物有一定抑制腫瘤細(xì)胞生長(zhǎng)作用。

Abstract [Objective] To study antitumor constituents from Hydrocotyle sibthorpioides. [Method] The constituents were isolated by chromatography and identified by NMR spectroscopic methods. The antitumor experiment in vitro, the MTT was used to screen constituents of Hydrocotyle sibthorpioides. [Result] Compounds 1-4 were isolated and identified. Each extracted parts of Hydrocotyle sibthorpioides all have a certain inhibition effect on LA795, the inhibition effect of Nbutanol extract in high concentration of 800μg/ml is the strongest, while the inhibition effects of petroleum ether and ethyl acetate extracts are relative weaker. [Conclusion] The 4 compounds all have a certain inhibition effect on the growth of tumor cells.

天胡荽為傘形科天胡荽屬(Hydrocotyle)多年生草本植物天胡荽(Hydrocotyle sibthorpioides Lam.)的全草,生于潮濕路旁、草地、山坡、墻腳、河畔和溪邊等,主要分布于江蘇、安徽、浙江和江西等地;朝鮮,日本,亞洲東南部和南部等地也有分布[1-2]。天胡荽有清熱利濕和退黃功用,治療肝膽濕熱之黃疸,可單用,若與茵陳、柴胡等通用,則利濕退黃效果更好。治下痢日久,濕熱不清,可與刺梨根、石榴皮等水煎服,以清熱利濕,澀腸止痢。濕熱蘊(yùn)結(jié)下焦,腎或膀胱結(jié)石,小便不利,甚或刺痛尿血者,可單用該品煎湯,或與石葦、海金砂等清熱利尿藥同用。其還有清肝祛風(fēng)和退翳明目之功效,可治肝經(jīng)風(fēng)火眼痛,翳膜攀睛,可將其揉塞鼻中,或與旱蓮草搗爛外敷[1-2]?,F(xiàn)代藥理研究表明,天胡荽具有抗腫瘤和抗HBsAg等作用[3]。筆者利用色譜手段對(duì)天胡荽進(jìn)行分離純化,應(yīng)用NMR方法鑒定結(jié)構(gòu),并進(jìn)行MTT法抗腫瘤試驗(yàn),以期為天胡荽的進(jìn)一步開發(fā)利用提供依據(jù)。
3 結(jié)論
天胡荽的各萃取部位對(duì)LA795細(xì)胞株有一定的抑制作用,其中以正丁醇層萃取物在高濃度800 μg/ml時(shí)的抑制作用較強(qiáng),而石油醚和乙酸乙酯萃取物的抑制作用相對(duì)較弱。單體化合物對(duì)LA795細(xì)胞株的抑制作用均較弱。
