一種快速鑒別甜菜夜蛾蛹及成蟲(chóng)雌雄的簡(jiǎn)易方法

描述了一種快速準(zhǔn)確區(qū)分甜菜夜蛾蛹及成蟲(chóng)雌雄的方法。甜菜夜蛾雌、雄蛹的主要區(qū)別是:雌蛹第8腹節(jié)腹面有一較短的縱裂縫,裂縫兩側(cè)平坦無(wú)突起;雄蛹第9腹節(jié)腹面有一長(zhǎng)的縱裂縫,裂縫兩側(cè)有半圓形瘤狀突起。成蟲(chóng)期雌、雄蛾主要區(qū)別是:雄蛾體色較淺,腹部狹長(zhǎng)且末端有一圈黃色長(zhǎng)毛簇;雌蛾體色較深,腹部末端渾圓,毛簇較短,生殖孔清晰可見(jiàn)。以此標(biāo)準(zhǔn)所建立的甜菜夜蛾雌雄蛹及成蟲(chóng)的快速鑒定方法,對(duì)于田間性比、預(yù)測(cè)和種群動(dòng)態(tài)以及人工飼養(yǎng)等具有重要作用。

甜菜夜蛾 Spodoptera exigua (Hübner),又名貪夜蛾、白菜褐夜蛾,隸屬于鱗翅目Lepidoptera夜蛾科Noctuidae灰翅夜蛾屬Spodopter,是一種世界性分布、以為害蔬菜為主的多食性害蟲(chóng)。在蔬菜中,受害最重的是十字花科蔬菜和豆科蔬菜(洪曉月等,2007)。迅速準(zhǔn)確鑒別甜菜夜蛾蛹和成蟲(chóng)雌雄性別,對(duì)于了解其田間性比和預(yù)測(cè)種群動(dòng)態(tài)具有重要作用。同時(shí),甜菜夜蛾作為重要的實(shí)驗(yàn)材料,被應(yīng)用于大量科研試驗(yàn)中,對(duì)其蛹及成蟲(chóng)性別的準(zhǔn)確鑒別將有助于以上各種研究實(shí)驗(yàn)的開(kāi)展。有關(guān)甜菜夜蛾蛹和成蟲(chóng)的外部形態(tài)特征已有報(bào)道(洪曉月等,2007;趙艦,2011),但關(guān)于甜菜夜蛾蛹和成蟲(chóng)雌雄性別的鑒別特征未見(jiàn)報(bào)道。李建勛等(2008)曾用產(chǎn)卵孔來(lái)鑒別甜菜夜蛾蛹的雌雄性別,但具體的鑒別特征未見(jiàn)報(bào)道。為此作者對(duì)甜菜夜蛾蛹及成蟲(chóng)雌雄的外部形態(tài)特征進(jìn)行了觀察,總結(jié)出了快速鑒別甜菜夜蛾蛹及成蟲(chóng)雌雄的鑒別方法。經(jīng)羽化后成蟲(chóng)鑒定結(jié)果,正確率為100%。
1.1 供試蟲(chóng)源
甜菜夜蛾蟲(chóng)源購(gòu)自中山大學(xué)昆蟲(chóng)研究所,并在本實(shí)驗(yàn)室連續(xù)飼養(yǎng),觀察鑒定。
1.2 試蟲(chóng)飼養(yǎng)方法
將甜菜夜蛾幼蟲(chóng)于人工氣候室內(nèi),置于一次性塑料杯中用人工飼料(楊建云等,2013)單頭飼養(yǎng)。幼蟲(chóng)老熟后,讓其在剩余的人工飼料中化蛹。幼蟲(chóng)化蛹后,把蛹放在簡(jiǎn)易雌雄蛹分辨盤(pán)上,在光學(xué)解剖鏡下進(jìn)行形態(tài)觀察和鑒定。簡(jiǎn)易雌雄蛹分辨盤(pán)的制作方法是:取一邊長(zhǎng)為12 cm正方形鐵板,將內(nèi)徑0.6 cm,外徑0.9 cm的硅膠管剪成長(zhǎng)度0.8 cm的小短塊,用膠槍圓圈狀固定于鐵板上,每板固定18-20個(gè)(圖1)。冷卻后備用。

將鑒定后的雌、雄蛹分別單頭放置于塑料杯里。甜菜夜蛾的蛹?xì)v期為5-6 d,初蛹為白色,隨著時(shí)間的延長(zhǎng)蛹顏色逐漸加深,變?yōu)辄S褐色。復(fù)眼位于頭部?jī)蓚?cè),黃褐色,羽化前變黑,后期蛹的顏色變深,即開(kāi)始羽化為成蟲(chóng)。待成蟲(chóng)羽化后,取將兩類成蟲(chóng)各一頭放于紙筒中,紙筒內(nèi)放置浸有10% 蜂蜜水的棉花團(tuán),且每天定時(shí)更換,供成蟲(chóng)取食補(bǔ)充營(yíng)養(yǎng)。羽化的成蟲(chóng)在光學(xué)顯微鏡下進(jìn)行形態(tài)觀察和鑒定。
試蟲(chóng)飼養(yǎng)條件為:溫度為27℃±1℃,相對(duì)濕度為70%-80%,光周期14L ∶10D(光照時(shí)間7 ∶00-21 ∶00)。
1.3 雌雄蛹的鑒別方法
取同一批次的甜菜夜蛾蛹,放于雌雄蛹簡(jiǎn)易分辨盤(pán)上,采用雙目解剖鏡(Motic K Series)觀察蛹。觀察甜菜夜蛾蛹的外部形態(tài)發(fā)現(xiàn),在蛹的第8、9節(jié)腹節(jié)腹面有一縱裂縫,根據(jù)此縱裂縫位置的不同,可將甜菜夜蛾蛹分成兩類:一類其縱裂縫位于第8腹節(jié)腹面(圖2,A);另外一類則位于第9腹節(jié)腹面,且位于第9腹節(jié)的縱裂縫兩側(cè)有兩個(gè)明顯的半圓形瘤狀突起(圖2,B)。以此標(biāo)準(zhǔn)將兩類蛹分開(kāi)羽化。羽化后,將兩類成蟲(chóng)一一配對(duì),并放進(jìn)紙筒內(nèi)觀察其交配和產(chǎn)卵,最終以產(chǎn)卵者視為雌成蟲(chóng)。
1.4 雌雄成蟲(chóng)的鑒別方法
收集羽化成蟲(chóng),觀察雌雄成蟲(chóng)的外部形態(tài)差異。用VHX-5000數(shù)碼顯微系統(tǒng)(KEYENCE 基恩士)對(duì)甜菜夜蛾蛹及成蟲(chóng)進(jìn)行形態(tài)拍照,并使用Photoshop CC(Adobe System, USA)進(jìn)行圖片的后期編輯(崔俊芝和葛斯琴,2012)。將鑒定出的新鮮雄蛾和雌蛾的腹部末端剪下,放入5% NaOH溶液中煮沸5-10 min,取出,在雙目解剖鏡下除去肌肉組織,將雄蛾的外生殖器直接制成標(biāo)本并用體式顯微鏡觀察、拍照。雌蛾的外生殖器經(jīng)染色后制成標(biāo)本用體式顯微鏡觀察、拍照。
2.1 雌雄蛹的外部形態(tài)特征分析
甜菜夜蛾蛹體長(zhǎng)7-12 cm。初蛹為白色,后逐漸變?yōu)辄S褐色,近羽化時(shí)顏色變深,復(fù)眼位于頭部?jī)蓚?cè),黃褐色,羽化前顏色變黑。蛹尾部末端有臀刺2根。雌蛹,第8腹節(jié)腹面近前緣有一短縱裂縫,長(zhǎng)度為100-200 μm,裂縫兩側(cè)平坦,無(wú)突起,裂縫前端為生殖孔,后端為產(chǎn)卵孔;裂縫離肛門(mén)的距離較遠(yuǎn),距離為500-800 μm;腹部末端鈍圓,第8-10腹節(jié)分節(jié)不明顯(如圖2,A)。雄蛹,第8腹節(jié)腹面無(wú)縱裂縫;第9腹節(jié)腹面中央有一縱裂縫,較長(zhǎng),長(zhǎng)度為200-300 μm,裂縫兩側(cè)各有一半圓形的瘤狀突起,縱裂離肛門(mén)的距離較近,距離為200-400 μm;腹部末端較尖,第8-10腹節(jié)分節(jié)較為明顯(如圖2,B)。

2.2 成蟲(chóng)雌雄的鑒定
將觀察區(qū)分的甜菜夜蛾蛹200頭(雄蛹100頭,雌蛹100頭)分別放置,待羽化后根據(jù)成蟲(chóng)形態(tài)特征對(duì)蛹期鏡檢的結(jié)果進(jìn)行分析。存活雄成蟲(chóng)85頭,雌成蟲(chóng)96頭,鑒定雄成蟲(chóng)85頭,雌成蟲(chóng)96頭,鑒別準(zhǔn)確率均為100%(見(jiàn)表1)。

2.3 雌雄成蟲(chóng)的外部特征
甜菜夜蛾成蟲(chóng)體長(zhǎng)10-14 mm,觸角絲狀。翅展19-25 mm。體灰褐色,前翅中央近前緣外方有腎形斑一個(gè),內(nèi)方有環(huán)形斑一個(gè),外緣有一列黑色三角形斑。后翅銀白色,略帶粉紅色,翅緣灰褐色。
雌蛾腹部末端圓筒形,有黃色毛簇,較短,生殖孔清晰可見(jiàn)。雄蛾腹部末端狹長(zhǎng),較尖,有一圈黃色長(zhǎng)毛簇(圖3)。雄蛾生殖器見(jiàn)圖4(A和B),雌蛾生殖器見(jiàn)圖3(C)。

甜菜夜蛾是世界性、爆發(fā)性害蟲(chóng)(洪曉月等,2007)。準(zhǔn)確區(qū)分甜菜夜蛾蛹和成蟲(chóng)雌雄性別,對(duì)該蟲(chóng)的發(fā)生及種群數(shù)量的預(yù)測(cè)預(yù)報(bào)工作有重要意義。本文觀察到甜菜夜蛾雌雄蛹的主要區(qū)別是:雌蛹第8腹節(jié)腹面有一較短的縱裂縫,縱裂縫兩側(cè)平坦無(wú)突起;雄蛹第9腹節(jié)腹面有一長(zhǎng)的縱裂縫,縱裂縫兩側(cè)均有半圓形瘤狀突起。就雌雄蛹腹部分節(jié)來(lái)看,甜菜夜蛾蛹與斜紋夜蛾Spodopteralitura(Fabricius)和黑腎卷群夜蛾P(guān)lecopteraoculata(Moore)相似(林威鵬等,2015;劉怡等,2015),均表現(xiàn)雄蛹8-10腹節(jié)分節(jié)較為明顯;而雌蛹8-10腹節(jié)分節(jié)不明顯;就縱裂縫來(lái)看,甜菜夜蛾雄蛹縱裂縫兩側(cè)有瘤狀突,雌蛹沒(méi)有瘤狀突,與旋幽夜蛾Scotogrammatrifolii(Rottemberg)和斜紋夜蛾類似(趙琦等,2011;林威鵬等,2015),與黑腎卷群夜蛾不同(劉怡等,2015);就位置來(lái)看,雄蛹生殖孔位于第9腹節(jié),雌蛹生殖孔位于第8腹節(jié),與桐花樹(shù)毛顎小卷蛾Lasiognathacellifera(Meyrick)、菜粉蝶Pierisrapae(Linnaeus)、小菜蛾P(guān)lutellaxylostella(Linnaeus)、二點(diǎn)委夜蛾Athelislepigone(Moschler)、團(tuán)花絹螟蛾Dinahaniaglauculalis(Guenée)、香梨優(yōu)斑螟蛾euzopheraPyriella(Yang)、嘴壺夜蛾Oraesiaemarginata(Fabricius)、柑橘長(zhǎng)卷蛾Homonacoffearia(Meyrick)、斜紋夜蛾等極其相似(李德偉等,2008;陳增良等,2010;陳藝欣等,2011;馬繼芳等,2011;劉志韜等,2014;張蒙等,2014;馮波等,2015;李莉玲等,2015;林威鵬等,2015)。甜菜夜蛾成蟲(chóng)雌雄差異則主要體現(xiàn)在體色及腹部末端外部形態(tài)上。雄蟲(chóng)體色較淡,且腹部較為狹長(zhǎng),末端生殖器外為黃色長(zhǎng)毛簇;而雌性體色較深,腹部渾圓,末端生殖器外為黃色短毛簇。上述甜菜夜蛾蛹和成蟲(chóng)雌雄性別的鑒別特征,可為甜菜夜蛾交配行為的觀察、性信息素的利用、成蟲(chóng)對(duì)揮發(fā)性物質(zhì)差異比較、電生理反應(yīng)等方面研究提供參考。

A rapid and accurate method for the sex recognition ofSpodopteraexiguaHübner (Lepidoptera: Noctuidae) pupae and adults was described in this paper. In female pupae, the 8thabdominal segment had a short lengthways crack in the middle with flat sides and no humps. In males, the 9thhad a central crack with adistinct semi-circular strumae each side. The male adult’s abdomen were long and narrow and had some cylindrical tail hair in the end, while the female adults were darker in color and their abdominal ends appeared round in shape, and a gonopore was obviously visible. The method is efficient for use in the field and has the potential to determining the sex ratio, predicting population trends and artificial feeding inS.Exigua.
