目的分析比較不同產(chǎn)地杜仲葉的紅外光譜,建立杜仲葉的薄層色譜鑒別方法,為其質(zhì)量控制提供依據(jù)。方法利用紅外光譜、二階導(dǎo)數(shù)譜圖對(duì)6種不同產(chǎn)地杜仲葉進(jìn)行特征峰的指認(rèn)和對(duì)比分析,并進(jìn)行聚類分析;通過薄層色譜對(duì)杜仲葉中活性成分進(jìn)行分離和鑒別。結(jié)果通過特征峰峰形、峰位與峰強(qiáng)的對(duì)比,發(fā)現(xiàn)不同產(chǎn)地杜仲葉之間存在差異,聚類結(jié)果顯示可將6個(gè)產(chǎn)地杜仲葉分為1類;薄層色譜結(jié)果顯示6批樣品主斑點(diǎn)一致,但不同產(chǎn)地的樣品間微量成分斑點(diǎn)存在差異。結(jié)論2種方法直觀、簡(jiǎn)單、方便、快速,為杜仲葉的分離鑒別和質(zhì)量控制提供了依據(jù)。

杜仲葉是杜仲科杜仲屬植物杜仲Eucommia ulmoides Oliv.的干燥葉[1]。杜仲葉與皮的化學(xué)成分基本一致[2],且具有同等功效,在醫(yī)藥、保健、食品方面發(fā)展前景廣闊。杜仲葉富含綠原酸、槲皮素、山柰酚、桃葉珊瑚苷、京尼平甘酸等活性物質(zhì)及多種氨基酸、維生素[3],有良好的降壓、利尿、保肝、抗腫瘤、抗菌、抗衰老和中樞鎮(zhèn)靜等多種藥理作用。本研究采用傅里葉變換紅外光譜[4]、二階導(dǎo)數(shù)紅外光譜、聚類分析[5-6]與薄層色譜法[7]對(duì)不同產(chǎn)地的6種杜仲葉進(jìn)行鑒別研究,為杜仲葉的鑒定和質(zhì)量控制提供依據(jù)。

1 儀器與試藥

TENSOR27傅里葉變換近紅外光譜儀(德國(guó)Bruker 公司)DTGS檢測(cè)器,光譜掃面范圍4000~400 cm-1,光譜分辨率2 cm-1,累計(jì)掃描次數(shù)32次。769YP-15A粉末壓片機(jī)(天津市科器高新技術(shù)公司);70-1型遠(yuǎn)紅外干燥箱(上海申光儀器儀表有限公司);薄層色譜點(diǎn)樣器(Linomat5 CAMAG),薄層色譜點(diǎn)樣針(CAMAG),薄層色譜照相儀(REPROSTAR 3),薄層色譜掃描儀(TLC SANNER3 CAMAG)

杜仲葉樣品采自5個(gè)不同地區(qū)(S1為2014年摘于新疆烏魯木齊,S2為2015年摘于新疆烏魯木齊,S3為陜西銅州,S4為湖北襄陽(yáng),S5為湖南張家界,

ObjectiveTo analyze and compare the Eucommia ulmoides leaves from different producing areas by IR; To establish a TLC method for identification of Eucommia ulmoides leaves; To provide a reference for its quality control.MethodsThe characteristic absorption peaks were identified and the positions and intensities of Eucommia ulmoides leaves from 6 producing areas were compared by IR and second derivative spectrum, including clustering analysis; Separation and identification of the active components in Eucommia ulmoides leaves were conducted by TLC.ResultsThere a difference exited in the characteristic absorption peaks in peakshapes, positions and intensities. And the samples from 6 producing areas could be divided into one category. Results found 6 batches of sample spots of consistent by TLC, but between samples of different producing areas trace components spots existed differences.ConclusionThe two methods are direct, simple, fast and convenient, which can provide a reliable evidence for identification and quality control of the Eucommia ulmoides leaves.
