桂木薯11號(hào)食用木薯新品種的選育

木薯(Manihot"esculenta"Crantz)為全球第六大糧食作物,廣泛種植于非洲、亞洲和南美洲熱帶地區(qū),是世界超過10億人的口糧,也是我國華南地區(qū)優(yōu)勢特色作物,全程不使用農(nóng)藥即可獲得高產(chǎn),具有低糖、低脂、富含膳食纖維的特點(diǎn),被譽(yù)為“淀粉之王”“地下糧倉”。近年來,世界糧食安全形勢日益嚴(yán)峻[1-4],《2022年全球糧食危機(jī)報(bào)告》顯示,2021年全球約1.93億人口面臨糧食危機(jī),較2020年人數(shù)增加26%[5]。據(jù)聯(lián)合國糧農(nóng)組織(FAO)統(tǒng)計(jì),世界木薯的產(chǎn)量呈持續(xù)平穩(wěn)增長趨勢,近年總產(chǎn)量3億t左右。我國是世界上最大的木薯進(jìn)口國,其中廣西種植面積和產(chǎn)量均占全國60%以上,但自2008年以來全國木薯種植面積和產(chǎn)量逐年下滑,貿(mào)易逆差持續(xù)擴(kuò)大[6-8],究其原因主要是木薯種植效益偏低導(dǎo)致。隨著人們對“大食物觀”及大健康產(chǎn)業(yè)的關(guān)注、木薯羹等加工產(chǎn)品火爆網(wǎng)絡(luò),木薯作為天然綠色的健康薯類雜糧開發(fā)利用,市場前景廣闊。因此,食用木薯新品種選育工作重要性日益突顯。

木薯遺傳背景復(fù)雜、基因組高度雜合,唯一栽培種的自然雜交群體中優(yōu)異資源稀有,重要性狀關(guān)鍵基因資源挖掘也嚴(yán)重滯后[9-11]。通過自交,一些控制重要性狀的基因的位點(diǎn)得以純合并產(chǎn)生加性效應(yīng),優(yōu)異表型性狀基因得以疊加表達(dá),為木薯品種選育和關(guān)鍵基因挖掘提供了重要途徑,國際上糯木薯、小顆粒木薯、各種抗性木薯等優(yōu)異資源都是在自交分離群體中篩選出來的[12-15],尚小紅等[16]對木薯自交群體也做了表型鑒定評價(jià)及分析,并從中選育出高類胡蘿卜素含量的木薯品種桂木薯9號(hào)[17]。同時(shí),氫氰酸含量、薯肉顏色是評價(jià)木薯品質(zhì)的重要指標(biāo),分子標(biāo)記輔助育種是品種選育的重要手段,通過與傳統(tǒng)選育種相結(jié)合,可有效提高木薯育種效率[18-20]。但是,目前國內(nèi)外對通過木薯自交群體選育優(yōu)新品種的研究較少,對分子標(biāo)記輔助育種的研究也主要集中在分子標(biāo)記開發(fā)方面,對其實(shí)際成功應(yīng)用的案例鮮有報(bào)道。

本研究以小芝麻木薯自交分離群體為材料,

輔助高鑒定準(zhǔn)確率的SNAP分子標(biāo)記技術(shù),選育出食用木薯新品種桂木薯11號(hào)。桂木薯11號(hào)的選育,是基于自交分離群體利用分子標(biāo)記技術(shù)輔助選育木薯新品種的又一成功案例,為今后品種選育和推動(dòng)產(chǎn)業(yè)轉(zhuǎn)型升級(jí)提供技術(shù)支撐和品種支撐。

1""材料與方法

1.1""材料

本研究選育的木薯品種,以國外引進(jìn)的小芝麻木薯品種為親本,以當(dāng)前我國主栽食用木薯品種華南9號(hào)為對照(CK)。以廣西農(nóng)業(yè)科學(xué)院南寧武鳴基地種植的木薯品種新選048及其自交所得的45份木薯種質(zhì),以及6個(gè)木薯品種的葉片為SNAP分子標(biāo)記材料。

1.2""方法

1.2.1""自交分離群體構(gòu)建與優(yōu)良株系篩選""2011年,在廣西農(nóng)業(yè)科學(xué)院南寧武鳴木薯育種基地對小芝麻木薯進(jìn)行自交授粉,輔助胚挽救技術(shù)獲得自交株系實(shí)生種。2012—2013年通過田間性狀測定輔助SNAP分子標(biāo)記[21-22]技術(shù)進(jìn)行單株評價(jià),選擇株型好、結(jié)薯性好、收獲指數(shù)高、鮮薯氫氰酸含量低、薯肉淺黃色的優(yōu)良株系,篩選出該種質(zhì),田間編號(hào)NK-10。

1.2.2""品種比較試驗(yàn)nbsp;"2014—2016年,在廣西農(nóng)業(yè)科學(xué)院南寧武鳴木薯育種基地進(jìn)行品種比較試驗(yàn),其中2014年進(jìn)行初級(jí)系比試驗(yàn),2015年進(jìn)行中級(jí)系比試驗(yàn),2016年進(jìn)行高級(jí)系比試驗(yàn),試驗(yàn)采用隨機(jī)區(qū)組設(shè)計(jì),每個(gè)小區(qū)面積為6.4"m′"4.0"m=25.6"m2,株行距為1.0"m′0.8"m,重復(fù)3次。測定性狀包括植物學(xué)特性、生物學(xué)特性、豐產(chǎn)性、氫氰酸含量、抗性。其中,氫氰酸含量采用曾文丹等[23]的快速檢測比色法,顯示顏色分為10個(gè)區(qū)域,對應(yīng)10個(gè)等級(jí),Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ、Ⅵ、Ⅶ、Ⅷ、Ⅸ、Ⅹ分別代表其氫氰酸含量區(qū)間為0~5、5~10、10~20、20~30、30~50、50~100、100~200、200~400、400~800、gt;800"mg/kg;抗性采用陸柳英等[24]的木薯朱砂葉螨抗性鑒定方法,評價(jià)標(biāo)準(zhǔn)見表1。

1.2.3""品種區(qū)域性試驗(yàn)""2017—2018年,分別在廣西南寧市武鳴區(qū)、廣西北海市合浦縣、廣西崇左市龍州縣、海南樂東黎族自治縣、江西贛州市贛縣區(qū)進(jìn)行品種區(qū)域性試驗(yàn),采用隨機(jī)區(qū)組設(shè)計(jì),每個(gè)小區(qū)面積為60"m2,株行距為1.0"m×0.8"m,重復(fù)3次。測定性狀包括適應(yīng)性、豐產(chǎn)性、氫氰酸含量、抗性。

1.2.4""品種生產(chǎn)性試驗(yàn)""2019—2020年,分別在廣西南寧市武鳴區(qū)、廣西北海市合浦縣、廣西崇左市龍州縣、海南樂東黎族自治縣、江西贛州市贛縣區(qū)進(jìn)行品種生產(chǎn)性試驗(yàn),參試品種采用間排法排列,重復(fù)3次,每個(gè)試驗(yàn)點(diǎn)木薯總種植面積分別為1.00、1.33、2.00、0.67、0.67"hm2。測定性狀包括適應(yīng)性、豐產(chǎn)穩(wěn)產(chǎn)性、氫氰酸含量、食用品質(zhì)。其中食用品質(zhì)評價(jià)方法采用8人評分小組對各品種蒸木薯的苦度、香度、甜度、黏度、面度、纖維度共6個(gè)指標(biāo)進(jìn)行評分,評分后按最終得分的平均分進(jìn)行質(zhì)量分級(jí)(表2)。

1.2.5""營養(yǎng)品質(zhì)測定""采用五點(diǎn)取樣法對桂木薯11號(hào)木薯鮮薯進(jìn)行取樣,均勻混樣后隨機(jī)取5"kg鮮薯樣品,委托廣西益譜檢測技術(shù)有限公司對鮮薯可溶性糖含量、粗纖維含量、最小干物質(zhì)含量、淀粉占干物質(zhì)比例、直鏈淀粉占干物質(zhì)比例、蛋白質(zhì)含量進(jìn)行測定;委托廣西壯族自治區(qū)分析測試中心對鮮薯氫氰酸含量、淀粉含量進(jìn)行測定。

1.2.6""指紋圖譜鑒定""采用陸柳英等[17]的方法利用34對微衛(wèi)星標(biāo)記引物通過13份木薯品種(種質(zhì))進(jìn)行引物篩選并擴(kuò)增獲得等位基因,通過等位基因?qū)Ρ全@取桂木薯11號(hào)唯一特征帶作為其標(biāo)記指紋。

2""結(jié)果與分析

2.1""品種比較試驗(yàn)結(jié)果

如表3所示,桂木薯11號(hào)3年平均鮮薯產(chǎn)量為41.88"t/hm2,較CK品種平均增產(chǎn)23.29%;氫氰酸含量為10~20"mg/kg,比CK品種氫氰酸含量(20~30"mg/kg)低一個(gè)等級(jí);中抗朱砂葉螨。

2.2""品種區(qū)域性試驗(yàn)結(jié)果

如表4所示,桂木薯11號(hào)在廣西、海南、江西品種表現(xiàn)穩(wěn)定,平均鮮薯產(chǎn)量為45.15"t/hm2,顯著高于CK,平均增產(chǎn)30.33%;氫氰酸含量為10~20"mg/kg,比CK低一個(gè)等級(jí);中抗朱砂葉螨。

2.3""品種生產(chǎn)性試驗(yàn)結(jié)果

如表5所示,桂木薯11號(hào)在廣西、海南、江西品種表現(xiàn)穩(wěn)定,平均鮮薯產(chǎn)量為44.80"t/hm2,顯著高于對照,平均增產(chǎn)24.07%;蒸木薯食用味品質(zhì)評價(jià)等級(jí)均為優(yōu),但桂木薯11號(hào)優(yōu)于華南9號(hào)。

2.4""營養(yǎng)品質(zhì)測定結(jié)果

2019年12月取樣木薯鮮薯送至廣西益譜檢測技術(shù)有限公司檢測,2021年12月取樣木薯鮮薯送至廣西壯族自治區(qū)分析測試中心檢測。檢測結(jié)果如表6所示,桂木薯11號(hào)比CK鮮薯可溶

性糖含量增加2.79%,粗纖維含量增加0.10%,淀粉含量降低2.10%,直鏈淀粉占干物質(zhì)比例降低1.84%,蛋白質(zhì)含量降低0.84%。

2.5""品種生物學(xué)特性

如圖1所示,桂木薯11號(hào)成熟期9~10個(gè)月,株型緊湊,株高2.0~3.5"m;葉片綠色,掌狀深裂,裂片7片,裂片披針形,中間裂片長12.0~20.0"cm,中間裂片寬3.5~6.0"cm,頂端嫩葉淺綠色,葉柄黃綠色,葉柄長20.0~30.0"cm,葉脈和嫩莖淺綠色;成熟主莖莖粗2.5~4.0"cm,中下部外皮灰白色或灰褐色,內(nèi)皮深綠色;結(jié)薯集中,掌狀平伸,薯形圓柱-圓錐形,塊根外皮褐色,內(nèi)皮乳黃色,薯肉淡黃色。圓錐花序,花萼淡黃色,子房綠色,"蒴果橢圓狀,果皮深綠色,成熟種子扁圓形,種皮黑色硬殼質(zhì),具花紋。

2.6""品種指紋圖譜

通過指紋圖譜鑒定,篩選出1對擴(kuò)增條帶清晰且多態(tài)性好的引物C107,其中,正向引物為5?-GAGCAACTAAGGAAGCCACA-3?,反向引物為5?-AACTAGCGGCACTCAAATCAAT-3?,連鎖群為8個(gè),連鎖群位置為43.238。擴(kuò)增后可以獲得5個(gè)等位基因(表7、圖2),桂木薯11號(hào)具有唯一的特征帶C107-e。C107可作為桂木薯11號(hào)的標(biāo)記指紋。

3""討論

桂木薯11號(hào)食用木薯新品種株型緊湊,塊根肉質(zhì)淺黃色,鮮薯薯肉氰化物含量為12.30"mg/kg,干物質(zhì)含量為38.40%,平均淀粉含量為28.20%,可溶性糖含量為4.71%,粗纖維含量為1.80%;鮮薯產(chǎn)量為43.35"t/hm2,中抗朱砂葉螨;鮮薯蒸煮食用口感香、軟、粉、糯,被譽(yù)為“香芋木薯”。同時(shí),采用相關(guān)性分析和主成分分析對桂木薯11號(hào)鮮薯加工適宜性進(jìn)行研究,結(jié)果表明其加工適宜性良好,可作為木薯羹、木薯汁、木薯薯片等加工原料[25-26]。2022年,桂木薯11號(hào)榮獲第三屆中國(廣西)-東盟現(xiàn)代種業(yè)發(fā)展大會(huì)“好種好品”金獎(jiǎng),并簽訂了125.00萬元的以桂木薯11號(hào)作為主要推廣品種的成果轉(zhuǎn)化合同。當(dāng)前,隨著人們對“大食物觀”及大健康產(chǎn)業(yè)的關(guān)注、木薯羹等加工產(chǎn)品火爆網(wǎng)絡(luò),木薯作為天然綠色的健康薯類雜糧開發(fā)利用,市場前景廣闊[27]。

桂木薯11號(hào)的選育,是基于自交分離群體利用分子標(biāo)記技術(shù)輔助選育木薯新品種的又一成功案例,證明以自交分離群體為材料選育木薯優(yōu)良新品種具有極大可行性。自交可使得一些重要性狀基因位點(diǎn)純合并產(chǎn)生加性效應(yīng),使一些控制優(yōu)異性狀的基因?qū)Ρ硇偷呢暙I(xiàn)值疊加[28]。KAWUKI等[29]、ROJAS等[30]、FREITAS等[31]以木薯自交系群體為材料,均篩選到了農(nóng)藝性狀顯著高于對應(yīng)親本的木薯品種(種質(zhì))。尚小紅等[16]研究發(fā)現(xiàn)新選048木薯自交分離群體出現(xiàn)了廣泛的性狀分離。陸柳英等[17]以新選048自交群體為材料,選育出薯肉深黃色、β-胡蘿卜素含量454"μg/100"g的桂木薯9號(hào)食用木薯品種。肖亮等[32]通過對國內(nèi)外木薯重要性狀基因研究進(jìn)展進(jìn)行分析,提出構(gòu)建自交分離群體和多組學(xué)聯(lián)合分析是挖掘木薯關(guān)鍵基因的重要手段。

但是,通過自交選育木薯優(yōu)新品種也有一定的局限性。首先,生產(chǎn)中優(yōu)異木薯品種通常不開花或不容易開花,因此很難獲得自交材料;其次,木薯是基因組高度雜合的二倍體,自交易出現(xiàn)衰退現(xiàn)象,獲得超親材料的概率比較低[9]。

4""結(jié)論

2021年12月,桂木薯11號(hào)食用木薯新品種通過由農(nóng)業(yè)農(nóng)村部南亞熱帶作物中心組織的現(xiàn)場鑒評,結(jié)果與歷年區(qū)域性試驗(yàn)和生產(chǎn)性試驗(yàn)結(jié)果相一致。2023年3月,桂木薯11號(hào)獲得熱帶作物品種審定證書,其外觀品質(zhì)和食用品質(zhì)優(yōu)良,種植經(jīng)濟(jì)效益高,加工適宜性好,適宜在廣西、廣東、海南、江西等木薯產(chǎn)區(qū)推廣。桂木薯11號(hào)的選育,是基于自交分離群體利用分子標(biāo)記技術(shù)輔助選育木薯新品種的又一成功案例,為推動(dòng)木薯產(chǎn)業(yè)轉(zhuǎn)型升級(jí)提供技術(shù)支撐和品種支撐。
