慈菇消脂丸對(duì)非酒精性脂肪性肝炎模型大鼠JNK1、p-c-Jun和Caspase-8表達(dá)的影響

摘要:目的 ?觀察慈菇消脂丸對(duì)非酒精性脂肪性肝炎模型大鼠JNK1、p-c-Jun、Caspase-8表達(dá)的影響,探討其相關(guān)作用機(jī)制。方法 ?實(shí)驗(yàn)大鼠隨機(jī)分為正常組、模型組、陽性對(duì)照組和慈菇消脂丸高、中、低劑量組,每組10只。采用高脂飼料喂養(yǎng)大鼠制作非酒精性脂肪性肝炎模型大鼠,造模同時(shí)給予相應(yīng)藥物干預(yù)。造模12周后處死全部大鼠,取出肝臟。電鏡觀察肝細(xì)胞超微結(jié)構(gòu),TUNEL法檢測(cè)肝細(xì)胞凋亡,qRT-PCR和Western blot分別檢測(cè)肝組織JNK1、p-c-Jun、Caspase-8 mRNA和蛋白的表達(dá)。結(jié)果 ?電鏡觀察顯示,模型組大鼠肝細(xì)胞腫脹呈氣球樣變,胞漿中布滿眾多大小不一的脂滴。與正常組比較,模型組大鼠肝細(xì)胞凋亡顯著增加,JNK1、p-c-Jun、Caspase-8 mRNA和蛋白表達(dá)顯著升高;與模型組比較,慈菇消脂丸各劑量組和陽性對(duì)照組大鼠肝細(xì)胞脂肪樣變顯著減少,細(xì)胞凋亡顯著下降,JNK1、p-c-Jun、Caspase-8 mRNA和蛋白表達(dá)顯著降低。結(jié)論??慈菇消脂丸通過調(diào)節(jié)JNK信號(hào)通路抑制肝細(xì)胞凋亡,對(duì)非酒精性脂肪性肝炎模型大鼠有一定的治療作用。

Effects of Cigu Xiaozhi?Pills on Expressions of JNK1, p-c-Jun and Caspase-8 in Non-alcoholic Steatohepatitis Model Rats

Abstract: Objective?To observe the effects of Cigu Xiaozhi?Pills on the expressions of JNK1, p-c-Jun and Caspase-8 in non-alcoholic steatohepatitis (NASH) model rats; To discuss its related mechanism of action. Methods?Totally sixty SPF grade male SD rats were divided into six groups randomly: normal control group, model group, positive medicine group, Cigu Xiaozhi?Pills high-, medium- and low-dosage groups, with 10 rats in each group. NASH model was established by feeding rats with high fat diet. At the same time, the model rats were given medicine intervention. At the end of 12 weeks, all the experimental rats were killed and the livers were taken. The ultrastructure of liver cells was observed by transmission electron microscope. Hepatocyte apoptosis was detected by TUNEL method. The expressions of JNK1, p-c-Jun, Caspase-8 mRNA and protein in liver tissue were detected by qRT-PCR and Western blot. Results?Under electron microscope, the liver cells in the model group were swollen in balloon shape, and the cytoplasm was covered with numerous fat droplets of different sizes. Compared with the normal group, the apoptosis of hepatocytes significantly increased, and the expressions of JNK1, p-c-Jun, Caspase-8 mRNA and protein significantly increased in the model group. Compared with the model group, the adipose samples of hepatocytes in each Cigu Xiaozhi Pills group and positive medicine group decreased significantly; the apoptosis rate decreased significantly; the expressions of JNK1, p-c-Jun, Caspase-8 mRNA and protein decreased significantly. Conclusion?Cigu Xiaozhi Pills can inhibit hepatocyte apoptosis by regulating the JNK signal pathway and play a therapeutic role in NASH.

非酒精性脂肪性肝炎(nonalcoholic steatohepatitis,NASH)是非酒精性脂肪性肝病的重要臨床病理類型之一,其組織學(xué)具有肝細(xì)胞脂肪變、氣球樣變性和點(diǎn)狀壞死及混合性炎性細(xì)胞浸潤(rùn)的特征,可伴Mallory小體和不同程度的肝纖維化。研究表明,10%~25%單純性脂肪肝(nonalcoholic fatty liver,NAFL)患者將進(jìn)行性發(fā)展為NASH。30%~40%NASH合并進(jìn)展性肝纖維化,10%~15%NASH患者可進(jìn)展為肝硬化,而9%~26%NASH相關(guān)肝硬化患者在隨訪4~10年內(nèi)死亡[1-2]。NASH發(fā)病機(jī)制復(fù)雜,涉及脂代謝紊亂、胰島素抵抗、肝脂質(zhì)過氧化增加和細(xì)胞因子異常等多個(gè)相互關(guān)聯(lián)的病理生理環(huán)節(jié)。研究發(fā)現(xiàn),細(xì)胞凋亡是NASH的突出特點(diǎn),肝細(xì)胞凋亡與脂肪肝嚴(yán)重程度密切相關(guān),肝臟脂肪變?cè)街?,炎癥反應(yīng)越重,肝細(xì)胞凋亡也越多[3]。前期臨床和實(shí)驗(yàn)研究均顯示,慈菇消脂丸在治療脂肪性肝病、肝纖維化、高脂血癥及肥胖等方面已取得較好的療效,明顯減輕NAFLD肝組織脂肪變性程度及炎癥反應(yīng),對(duì)NAFLD具有確切的干預(yù)效應(yīng)[4-5]。本實(shí)驗(yàn)觀察慈菇消脂丸對(duì)NASH模型大鼠JNK1、p-c-Jun、Caspase-8表達(dá)的影響,探討其相關(guān)作用機(jī)制。

1 ?實(shí)驗(yàn)材料

1.1 ?動(dòng)物

SPF級(jí)雄性健康SD大鼠60只,8周齡,體質(zhì)量(200±20)g,甘肅中醫(yī)藥大學(xué)動(dòng)物實(shí)驗(yàn)中心提供,動(dòng)物許可證號(hào)SCXK(甘)2015-0002。飼養(yǎng)于溫度(22±2)℃、相對(duì)濕度50%~60%環(huán)境,12 h明暗交替照明。

1.2 ?藥物及制備

慈菇消脂丸(山慈菇10 g、法半夏15 g、土鱉蟲20 g、茯苓20 g、柴胡12 g、丹參20 g、薏苡仁30 g、黃芩10 g、澤瀉15 g、枸杞子20 g、山楂20 g等),飲片由甘肅中醫(yī)藥大學(xué)附屬醫(yī)院制劑中心提供。飲片用2 L冷蒸餾水浸泡30 min后,武火煮沸,改用文火保持沸騰煎煮30 min,過濾后,藥渣再加1.5 L冷蒸餾水煎煮1次,煮沸后保持沸騰30 min,過濾,合并2次濾液,放置過夜,離心,去除沉淀,電爐上用蒸發(fā)皿濃縮上清液,濃縮成3.6 g/mL為高劑量,稀釋1倍(1.8 g/mL)為中劑量,稀釋2倍(0.9 g/mL)為低劑量,4 ℃冰箱保存?zhèn)溆?。JNK抑制劑(SP600125,美國(guó)Santa Cruz公司)按0.5 mg/kg劑量用10 mL/L DMSO溶解,按5 mL/kg腹腔注射。

1.3 ?主要試劑與儀器

DMSO,美國(guó)Santa Cruz公司;Mab to JNK1抗體,美國(guó)Abcam公司,批號(hào)GR114659-3;Pab to JNK1抗體,美國(guó)Abcam公司,批號(hào)GR45117-7;p-c-Jun抗體,美國(guó)Abcam公司,批號(hào)GR3190868-1;Caspase 8抗體,美國(guó)GeneTex公司,批號(hào)Lot No.40044;β-actin,北京中杉金橋公司;BCA蛋白濃度測(cè)定試劑盒,Solarbio公司,批號(hào)Lot No.20170802;PrimeScriptTMRT Master Mix (Perfect Real Time)、TB GreenTMPremix Ex TaqTMⅡ(Tli RNaseH Plus)、TaKaRa MiniBEST Plant RNA Extraction Kit,TaKaRa公司。實(shí)時(shí)定量Applied Biosystems Q5儀,美國(guó)Thermo Fisher Scientific公司;Thermo-NANO核酸濃度測(cè)量?jī)x,上海創(chuàng)萌生物科技有限公司;Western blot轉(zhuǎn)膜儀、電泳儀,Bio-rad公司;酶標(biāo)儀,Thermo公司;HITACHI-HS500透射電鏡,日本HITACHI公司。

2 ?實(shí)驗(yàn)方法

2.1 ?分組

60只大鼠標(biāo)準(zhǔn)食水適應(yīng)性喂養(yǎng)7 d,隨機(jī)分為正常組、模型組、陽性對(duì)照組和慈菇消脂丸高、中、低劑量組,每組10只。

2.2 ?造模和給藥

參照文獻(xiàn)[6]方法,采用高脂飼料喂養(yǎng)制作模型。正常組給予普通標(biāo)準(zhǔn)飼料,同時(shí)按10 mL/kg鼠質(zhì)量灌服生理鹽水,每日1次;模型組給予高脂飼料喂養(yǎng),同時(shí)按10 mL/kg鼠質(zhì)量灌服生理鹽水,每日1次;慈菇消脂丸各劑量組在高脂飼料喂養(yǎng)的同時(shí)給予10 mL/kg體質(zhì)量相應(yīng)濃度藥液灌胃,每日1次;陽性對(duì)照組在高脂飼料喂養(yǎng)的同時(shí),第9周開始按5 mL/kg腹腔注射SP600125溶液,1次/d,連續(xù)4周。動(dòng)物給藥劑量按人與動(dòng)物體表面積比折算成等效劑量。實(shí)驗(yàn)持續(xù)至12周末結(jié)束。大鼠禁食24 h,次日10%水合氯醛腹腔注射麻醉,迅速取出肝臟。一塊置于4%多聚甲醛液中固定,石蠟包埋;一塊經(jīng)冷生理鹽水清洗后鋁箔包裹置于液氮內(nèi)速凍,-80 ℃冰箱保存?zhèn)溆谩?/p>

2.3 ?觀察指標(biāo)

2.3.1 ?電鏡觀察

取肝組織塊切成約1 mm3,用配好的電鏡固定液固定,漂洗,逐級(jí)脫水、浸透、固化、LKB型超薄切片機(jī)切片50~60 nm,枸櫞酸鉛染色,透射電鏡觀察肝細(xì)胞及細(xì)胞外基質(zhì)超微結(jié)構(gòu)。
