[摘要]目的 測定白細(xì)胞介素-21(IL-21)和CC趨化因子配體20(CCL20)在2,4-二硝基氯苯(DNCB)誘導(dǎo)的特應(yīng)性皮炎小鼠模型中的表達(dá)情況,探討二者在特應(yīng)性皮炎發(fā)病中的作用。方法 將12只BALB/c雌性小鼠,隨機(jī)分為模型組和對(duì)照組,每組6只,使用DNCB構(gòu)建小鼠特應(yīng)性皮炎模型。造模成功后采用酶聯(lián)免疫吸附法檢測小鼠血清中IL-21和CCL20的水平。結(jié)果 特應(yīng)性皮炎模型組小鼠血清中IL-21和CCL20的表達(dá)水平均高于對(duì)照組,差異有統(tǒng)計(jì)學(xué)意義(P<0.05),并且二者表達(dá)成正相關(guān)(r=0.59,P<0.05)。結(jié)論 IL-21和CCL20在特應(yīng)性皮炎小鼠模型中表達(dá)升高,且成正相關(guān),可能協(xié)同參與了特應(yīng)性皮炎的發(fā)生發(fā)展。

[Abstract] Objective To detect the expression of interleukin-21 (IL-21) and CC chemokine ligand 20 (CCL20) in 2,4-dinitrochlorobenzene (DNCB) induced mice models with atopic dermatitis and explore their roles in the pathogenesis of atopic dermatitis. Methods Twelve female BALB/c mice were randomly divided into the model group and control group(6 in each). Mice models with atopic dermatitis were established by DNCB. After successful modeling, the serum levels of IL-21 and CCL20 were detected by enzyme-linked immunosorbent assay(ELISA). Results The serum levels of IL-21 and CCL20 in the model group with atopic dermatitis were significantly higher than those in the control group (P<0.05). There was a positive correlation between the serum levels of IL-21 and CCL20 in the model group(r=0.59, P<0.05).Conclusions The expression of IL-21 and CCL20 in the mouse model of atopic dermatitis is increased and positively correlated, which may be synergistically involved in the occurrence and development of atopic dermatitis.

特應(yīng)性皮炎(AD)是由多種免疫、炎性細(xì)胞和細(xì)胞因子參與的慢性炎癥性皮膚病,皮損頑固難愈,病因和發(fā)病機(jī)制較為復(fù)雜,與遺傳、免疫異常、皮膚屏障受損、生存環(huán)境和感染等因素有關(guān)[1]。不同亞型CD4+T細(xì)胞介導(dǎo)的免疫失衡是AD發(fā)病的重要環(huán)節(jié)[2-3]。白細(xì)胞介素-21(IL-21)是IL-2家族中重要的多效能細(xì)胞因子,可調(diào)節(jié)淋巴細(xì)胞和自然殺傷(NK)細(xì)胞的增殖分化,廣泛參與體內(nèi)免疫應(yīng)答和炎性反應(yīng)。CC趨化因子配體-20(CCL20)是CC亞族的趨化因子,可趨化表達(dá)CC趨化因子受體(CCR)6的淋巴細(xì)胞和未成熟樹突狀細(xì)胞向抗原局部定向遷移,與多種自身免疫性疾病有關(guān)。本研究通過測定AD小鼠模型血清中IL-21和CCL20的表達(dá)變化,探討二者在AD發(fā)病中的作用。
1材料與方法
1.1實(shí)驗(yàn)動(dòng)物與主要試劑
無特定病原體動(dòng)物(SPF)級(jí)7~8周齡健康BALB/c雌性小鼠12只,動(dòng)物合格證號(hào)SCXK(京)2015-0006,由山西大同大學(xué)動(dòng)物實(shí)驗(yàn)中心提供。2,4-二硝基氯苯(DNCB)購自美國Sigma公司,小鼠IL-21、CCL20酶聯(lián)免疫吸附法(ELISA)檢測試劑盒購自武漢伊萊瑞特生物科技有限公司。本研究已通過本院醫(yī)學(xué)倫理委員會(huì)的審核批準(zhǔn)。
1.2方法
1.2.1實(shí)驗(yàn)動(dòng)物造模? 12只BALB/c小鼠清潔級(jí)實(shí)驗(yàn)室飼養(yǎng)1周,適應(yīng)環(huán)境后開始實(shí)驗(yàn),隨機(jī)分為模型組和對(duì)照組,各6只。參考文獻(xiàn)[4-5]構(gòu)建AD小鼠模型,實(shí)驗(yàn)前剃除小鼠背部毛發(fā),面積約3 cm×2 cm。模型組小鼠第1~3天背部剃毛區(qū)外涂100 μl 1%DNCB溶液(采用丙酮∶橄欖油=1∶3作為基質(zhì)配制),第4天繼續(xù)外涂100 μl 0.1%DNCB溶液,改為每間隔1 d外涂1次,直到第28天涂抹后造模完成。造模后小鼠背部皮膚增厚,可見明顯紅斑、丘疹、丘皰疹、滲出、堆積大量痂屑。對(duì)照組小鼠背部剃毛區(qū)僅外涂基質(zhì),涂抹劑量、間隔時(shí)間與模型組相同。
1.2.2皮膚組織病理檢查? 造模后,無菌條件下兩組小鼠背部剃毛區(qū)皮膚組織取材,10%甲醛溶液固定、脫水、透明、石蠟包埋、切片染色、封片,鏡下觀察兩組小鼠皮膚組織病理改變。
