摘 要: 隨著飼料中抗生素類飼料添加劑的全面禁用,中草藥飼料添加劑逐漸成為畜禽生產(chǎn)中替代抗生素的潛在產(chǎn)品。而中草藥經(jīng)過微生物發(fā)酵后,不僅能增加活性物質(zhì)含量和中草藥藥效,還能產(chǎn)生新的活性成分,降低中草藥毒副作用。將發(fā)酵中草藥飼料添加劑添加到畜禽飼料中能夠提高畜禽生產(chǎn)性能、改善健康狀況、提升畜禽產(chǎn)品品質(zhì)。本文歸納了發(fā)酵中草藥的優(yōu)點(diǎn),討論了發(fā)酵中草藥飼料添加劑對(duì)動(dòng)物生產(chǎn)的影響,并總結(jié)了中草藥發(fā)酵飼料添加劑使用中存在的問題,以期為發(fā)酵中草藥飼料添加劑在畜禽養(yǎng)殖中的研究和應(yīng)用提供參考依據(jù)。

Abstract:" With the comprehensive prohibition of antibiotic feed additives in livestock production, Chinese herbal feed additives have gradually become the potential alternative antibiotics. After microbial fermentation, Chinese herbal medicine not only can increase the content of active substances and enhance the efficacy, but also produce new active ingredients and reduce the toxic side effects of Chinese herbal medicine. Supplementation of fermented Chinese herbal medicine can improve the production performance, alleviate the health statues, and enhance the products quality of livestock animals. This review summarized the characteristics of the fermented Chinese herbal medicines, discussed the influence of fermented Chinese herbal feed additive on animal production, and explored the problems existing in its application. Hoping to provide reference for the research and application of fermented Chinese herbal feed additives in livestock and poultry production.

我國中草藥資源種類繁多,現(xiàn)有將近12 000種藥用植物資源可以作為中藥飼料添加劑的研發(fā)基礎(chǔ)。它們經(jīng)過合理的配伍和使用,在生產(chǎn)中具有多種生物學(xué)功效[1]。中草藥飼料添加劑是基于我國傳統(tǒng)中獸醫(yī)理論和實(shí)踐經(jīng)驗(yàn)開發(fā)的動(dòng)物飼料添加劑。它是以中草藥為主要原料,選取適宜的中草藥成分添加到動(dòng)物飼料中[2],能起到提高生產(chǎn)性能、促進(jìn)免疫調(diào)節(jié)、改善畜產(chǎn)品品質(zhì)等作用,相較于其他添加劑有著安全無殘留的特性,在生產(chǎn)中能夠起到關(guān)鍵的調(diào)控作用[3]。隨著人們對(duì)生態(tài)環(huán)境和畜產(chǎn)品質(zhì)量安全問題的重視程度逐漸加深,尤其是畜禽養(yǎng)殖過程中抗生素類添加劑被濫用的現(xiàn)象受到關(guān)注。國家農(nóng)業(yè)農(nóng)村部發(fā)布的194號(hào)公告中,我國已明確規(guī)定,飼料生產(chǎn)企業(yè)停止生產(chǎn)含有抗生素類飼料添加劑的商品飼料。近年來,中草藥作為畜禽生產(chǎn)中唯一被允許添加的促生長類添加劑受到廣泛關(guān)注,成為在養(yǎng)殖生產(chǎn)中替代抗生素的有效策略之一。
然而,大多數(shù)中草藥是植物源性成分,植物細(xì)胞壁致密的結(jié)構(gòu)阻礙了其活性物質(zhì)的釋放,再加上中草藥本身有效成分復(fù)雜且含量低,極大削弱了其作用的發(fā)揮。在4 000多年前,我國就開始將發(fā)酵技術(shù)應(yīng)用于中藥的炮制過程中,最初的發(fā)酵中藥主要是曲類中藥,例如半夏曲、六神曲、沉香曲等[4]。近年來,在現(xiàn)代微生物研究技術(shù)的發(fā)展下,中草藥的發(fā)酵過程在傳統(tǒng)發(fā)酵技術(shù)延伸下轉(zhuǎn)變?yōu)槔梦⑸锏霓D(zhuǎn)化作用以及微生物與中草藥之間的協(xié)同作用,實(shí)現(xiàn)更高效的炮制過程[5]。中草藥通過發(fā)酵能提高活性物質(zhì)含量、產(chǎn)生新的活性成分、降低有毒有害物質(zhì)成分等?,F(xiàn)代發(fā)酵技術(shù)的快速發(fā)展為中草藥飼料資源的開發(fā)利用注入了新的動(dòng)力,也使得發(fā)酵中草藥飼料添加劑在動(dòng)物生產(chǎn)中備受關(guān)注,且具有廣闊的開發(fā)前景,為養(yǎng)殖業(yè)的可持續(xù)發(fā)展提供了新的突破口。
1 發(fā)酵中草藥的優(yōu)點(diǎn)
1.1 提高活性物質(zhì)含量,增強(qiáng)藥效
在中草藥發(fā)酵過程中,微生物通過發(fā)酵產(chǎn)生的酶類物質(zhì)能夠溶解中草藥植物細(xì)胞壁,促進(jìn)壁內(nèi)有效成分的釋放,提高活性物質(zhì)的生物利用率。有研究發(fā)現(xiàn),通過利用胞外產(chǎn)丹寧酶真菌米曲霉對(duì)益母草、黃芪、當(dāng)歸等中草藥進(jìn)行固態(tài)發(fā)酵,總黃酮、多糖及總生物堿的平均含量顯著升高,這表明該中草藥經(jīng)發(fā)酵后活性物質(zhì)的釋放明顯上升[6]。以靈芝為藥用真菌,發(fā)酵雷公藤可獲得特定的靈雷菌質(zhì),通過對(duì)靈雷菌質(zhì)進(jìn)行急性毒性和藥效學(xué)試驗(yàn)研究,發(fā)現(xiàn)靈雷菌質(zhì)保持了原雷公藤藥材的藥效,但毒性相比于原雷公藤大為降低[7]。將丙冬甘寧中藥方利用芽胞桿菌進(jìn)行發(fā)酵,并用高效液相色譜法(HPLC)測(cè)定其有效物質(zhì)含量,發(fā)現(xiàn)發(fā)酵后甘草次酸、黃芩素含量升高,這表明經(jīng)發(fā)酵后丙冬甘寧中藥方劑中的活性物質(zhì)含量提高,有助于藥效增強(qiáng)[8]。用釀酒酵母發(fā)酵葛根芩連湯,觀察到總黃酮和總生物堿含量顯著增加,其作用效果顯著加強(qiáng)[9]。此外,以人參為發(fā)酵基質(zhì)進(jìn)行發(fā)酵,發(fā)現(xiàn)發(fā)酵后的人參勻漿SOD活性顯著升高,這表明乳桿菌發(fā)酵有助于增強(qiáng)人參抗氧化活性物質(zhì)的含量[10]。
中草藥渣是中草藥經(jīng)過加工提取有效成分后剩余的殘?jiān)糠郑ㄟ^檢測(cè)發(fā)現(xiàn),這些藥渣中仍含有大量不易被提取的纖維素、木質(zhì)素等成分[11]。近年來,中草藥渣潛在價(jià)值受到關(guān)注。有研究發(fā)現(xiàn),中草藥渣中還含有約30%的有效活性成分,經(jīng)過發(fā)酵處理能夠用于促進(jìn)畜禽生長,調(diào)節(jié)機(jī)體免疫力等[12]。丹參藥渣經(jīng)冬蟲草、靈芝、北蟲草等多菌種混合發(fā)酵后,可溶性多糖、真蛋白含量均顯著增加,這表明發(fā)酵中草藥渣可以提高活性物質(zhì)含量,提高發(fā)酵中藥渣營養(yǎng)價(jià)值[13]。將發(fā)酵后的甘草、黃芪等中草藥剩余殘?jiān)c小尾寒羊的日糧混合飼喂,觀察到小尾寒羊日增重、飼料轉(zhuǎn)化率均顯著增加,說明中草藥提取殘?jiān)l(fā)酵后仍具備一定的營養(yǎng)和利用價(jià)值[14]。
1.2 產(chǎn)生新的活性成分
借助微生物發(fā)酵的技術(shù),中草藥中的有效成分或微生物在發(fā)酵過程中產(chǎn)生的代謝物能夠作為起始物質(zhì),通過微生物轉(zhuǎn)化使中草藥產(chǎn)生新的活性化合物。用HPLC法對(duì)發(fā)酵后中草藥制劑活性成分進(jìn)行分析,發(fā)現(xiàn)出現(xiàn)了新的特征吸收峰,這表明發(fā)酵后的中草藥很可能生成了新的活性物質(zhì)[15-16]。人參、遠(yuǎn)志、桔梗和甘草等中草藥中主要有效成分都富含皂苷,經(jīng)過發(fā)酵處理,這些皂苷能夠轉(zhuǎn)化為葡萄糖苷、人參皂苷等新型活性物質(zhì)。通過將枳殼飲片進(jìn)行發(fā)酵,利用超高效液相色譜法(UPLC)進(jìn)行分析,觀察到出現(xiàn)兩個(gè)新的特征吸收峰,這兩個(gè)峰對(duì)應(yīng)的化合物分別是圣草酚-7-O-葡萄糖苷和橙皮素-7-O-葡萄糖苷,是兩種新的葡萄糖苷成分[17]。運(yùn)用無柄靈芝菌對(duì)黨參進(jìn)行發(fā)酵,發(fā)現(xiàn)產(chǎn)生了新的成分2,5-二甲醇呋喃[18]。利用枯草芽胞桿菌對(duì)中草藥三七根發(fā)酵,發(fā)現(xiàn)生成新的化合物人參皂苷Rh4[19]。以上研究均表明,中草藥經(jīng)過發(fā)酵后可以產(chǎn)生新的活性成分。
1.3 降低中草藥毒副作用
部分中草藥含有毒性成分,限制了其在畜禽生產(chǎn)中的應(yīng)用。發(fā)酵是減少中草藥毒性成分、副作用及不良反應(yīng)的有效途徑[20]。草烏具有散寒止痛的功效,由于其具有極高的毒性,使用時(shí)極易中毒且有可能喪命。運(yùn)用HPLC法對(duì)藥用真菌發(fā)酵的草烏進(jìn)行成分分析,發(fā)現(xiàn)其成分中劇毒的烏頭堿、中烏頭堿和次烏頭堿的含量都明顯降低,這表明草烏發(fā)酵后可以有效降低其毒副作用[21]。川烏具有抗炎鎮(zhèn)痛的作用,對(duì)風(fēng)濕病、類風(fēng)濕性關(guān)節(jié)炎都具有治療功效,但其本身也存在明顯毒性,臨床屢有用藥不當(dāng)致患者死亡的報(bào)道。有研究證明,利用靈芝菌發(fā)酵生川烏,生川烏中總生物堿和酯型生物堿含量顯著降低,有效減少生川烏毒性成分的作用[22]。將川烏、半夏、天南星等中草藥組成的化風(fēng)丹藥母進(jìn)行發(fā)酵,觀察到烏頭堿、甲烏頭堿、亞甲烏頭堿含量均顯著降低,這表明中草藥經(jīng)發(fā)酵后通過降低毒性成分含量,可有效減輕化風(fēng)丹藥母的毒副作用[23]。馬錢子是一種應(yīng)用廣泛的中草藥,擁有疏通脈絡(luò)、消腫止痛等功效,同時(shí)也具有強(qiáng)烈的毒性。因此,在使用前必須對(duì)其進(jìn)行炮制,使其減毒增效。利用朱紅栓菌固態(tài)發(fā)酵馬錢子,其發(fā)酵品的半數(shù)致死量(LD50)顯著升高[24]。同樣,將真菌發(fā)酵處理的馬錢子飼喂小鼠,小鼠的急性毒性LD50顯著升高,可見馬錢子經(jīng)發(fā)酵后毒性成分有效降解,可有效降低其毒副作用[25]。清風(fēng)藤是一種廣泛用于祛風(fēng)濕、通經(jīng)絡(luò)的中草藥,但該藥材存在減低白細(xì)胞的風(fēng)險(xiǎn)。通過7種食藥用真菌固體發(fā)酵小花清風(fēng)藤,發(fā)現(xiàn)食藥用真菌對(duì)小花清風(fēng)藤苯酚類物質(zhì)有顯著的降解作用,其中福州靈芝、靈芝EN2、平菇BP3等真菌能夠?qū)⑶屣L(fēng)藤中的毒性成分顯著降低或轉(zhuǎn)化為毒性較小乃至無毒的成分,這表明發(fā)酵中草藥對(duì)藥物毒性的降解具有顯著作用[26]。
2 發(fā)酵中草藥飼料添加劑對(duì)畜禽生產(chǎn)的影響
通過對(duì)中草藥進(jìn)行發(fā)酵處理,可有效提高活性物質(zhì)的含量,進(jìn)一步增強(qiáng)中草藥的藥效,并能產(chǎn)生新的活性物質(zhì)和降低部分有毒中草藥毒副作用。在畜禽養(yǎng)殖中添加可以促進(jìn)動(dòng)物生長性能、改善健康狀況、提高畜產(chǎn)品品質(zhì),是一種具有廣泛應(yīng)用潛力的飼料添加劑。
2.1 對(duì)畜禽生產(chǎn)性能的影響
發(fā)酵中草藥飼料添加劑可以用來補(bǔ)充和替代飼料中的營養(yǎng)成分,它能夠促進(jìn)消化酶的產(chǎn)生,改善飼料的適口性,進(jìn)而提高動(dòng)物的采食量,提升畜禽的生產(chǎn)性能。大量研究證實(shí),在豬場(chǎng)養(yǎng)殖生產(chǎn)中,使用發(fā)酵中草藥飼料添加劑可以提高仔豬的采食量,降低料肉比,改善育肥豬的屠宰性能,同時(shí)也能夠?qū)δ肛i的繁殖性能產(chǎn)生影響。在斷奶仔豬飼糧中添加乳酸菌混合發(fā)酵的五味子、車前子等中草藥或乳酸菌、芽胞桿菌等發(fā)酵后的黨參、黃芪、茯苓等中草藥配方,均顯著提高斷奶仔豬的平均日采食量,降低料肉比[27-28]。將黨參、黃芪、柴胡等中草藥發(fā)酵后加入到育肥豬的飼糧中,觀察到育肥豬的平均日增重和平均日采食量均提高,料肉比降低[29]。同樣,在育肥豬日糧中添加經(jīng)枯草芽胞桿菌、酵母菌液體發(fā)酵的中草藥,發(fā)現(xiàn)育肥豬的平均日增重顯著提高[30]。在育肥豬日糧中添加經(jīng)蟲草真菌液體發(fā)酵的復(fù)方中草藥制劑,育肥豬的屠宰率、瘦肉率有所提高,背膘厚明顯降低,屠宰性能及肉品質(zhì)顯著改善[31]。此外,將雙參發(fā)酵粉添加到妊娠母豬飼料中,與未添加的對(duì)照組相比,母豬啟動(dòng)分娩超過預(yù)產(chǎn)期的天數(shù)、產(chǎn)程時(shí)間、產(chǎn)仔間隔時(shí)間均顯著縮短,同時(shí)仔豬的平均日增重、初生重和斷奶重都顯著增加,可以看出發(fā)酵中草藥在顯著改善母豬繁殖性能的同時(shí),仔豬的生產(chǎn)性能也得到了提升[32]。
通過在家禽飼料中添加經(jīng)過發(fā)酵處理的中草藥,可以顯著改善飼料的適口性,促進(jìn)家禽的食欲,進(jìn)一步提高家禽的采食量,從而有效提升產(chǎn)蛋率和蛋均重,這種方法對(duì)于提高家禽的生產(chǎn)性能具有促進(jìn)作用。在京紅蛋雞產(chǎn)蛋后期母雞日糧中添加枯草芽胞桿菌發(fā)酵的黨參、白術(shù)等中草藥,發(fā)現(xiàn)母雞產(chǎn)蛋率顯著提高,料蛋比顯著下降,總產(chǎn)蛋重上升[33]。進(jìn)一步研究發(fā)現(xiàn),飼糧中添加發(fā)酵中草藥可以通過調(diào)控miRNA及其靶基因的表達(dá),參與營養(yǎng)物質(zhì)的調(diào)控和脂質(zhì)代謝,并影響生產(chǎn)性能[33]。選取20多種中草藥組成3種復(fù)合配方分為三組飼喂817肉雞,發(fā)現(xiàn)第一組發(fā)酵中草藥復(fù)合配方組末質(zhì)量、日增重質(zhì)量、料肉比相較對(duì)照組均顯著升高,肉雞生長性能明顯得到改善[34]。此外,通過在21日齡天府肉鴨的日糧中添加枯草芽胞桿菌和檸檬酸桿菌發(fā)酵的中草藥,肉鴨的末重、平均日增重、平均日采食量、屠宰率等指標(biāo)均有顯著提高,料重比顯著降低,明顯提高了肉鴨的生長性能[35]。
在反芻動(dòng)物日糧中添加經(jīng)發(fā)酵的中草藥飼料添加劑,可以通過增加反芻動(dòng)物采食量,從而提高產(chǎn)奶量和日增重等指標(biāo),促進(jìn)其生產(chǎn)性能的提升。將發(fā)酵黃芪添加到泌乳荷斯坦奶牛飼糧中,與未發(fā)酵組相比,奶牛平均日采食量和平均日產(chǎn)奶量顯著升高[36]。利用釀酒酵母菌和枯草芽胞桿菌對(duì)中草藥進(jìn)行發(fā)酵后,添加到熱應(yīng)激的荷斯坦奶牛的飼糧中,發(fā)現(xiàn)奶牛采食量顯著增多,產(chǎn)奶量顯著提高,改善了奶牛的生產(chǎn)性能[37]。以上結(jié)果均表明中草藥經(jīng)發(fā)酵后對(duì)反芻動(dòng)物的生產(chǎn)性能有明顯促進(jìn)作用。
2.2 對(duì)畜禽健康狀況的影響
中草藥經(jīng)發(fā)酵后添加到畜禽飼料中,可以改善畜禽的患病率,提高畜禽健康水平。研究發(fā)現(xiàn),將復(fù)方杜仲中草藥或板藍(lán)根、黃芪、黃芩等中草藥混勻后發(fā)酵添加到仔豬日糧中,發(fā)現(xiàn)發(fā)酵后中草藥飼料添加劑均可顯著降低仔豬腹瀉發(fā)生率和發(fā)病死亡率[38-39]。利用發(fā)酵復(fù)方中草藥制劑飼喂圍產(chǎn)期湖羊母羊,與空白組相比,顯著減少了母羊患圍產(chǎn)期相關(guān)疾病率[40]。在海蘭灰蛋雞飼糧中加入經(jīng)枯草芽胞桿菌發(fā)酵處理的黨參、白術(shù)、茯苓等中草藥,蛋雞的禽流感和新城疫的抗體水平有效提升,改善了禽類的抗病能力[41]。這些研究均表明發(fā)酵中草藥制劑可以有效促進(jìn)動(dòng)物健康水平。進(jìn)一步的研究發(fā)現(xiàn),在家禽健康狀況上發(fā)揮抗病功能的原因,可能是由于對(duì)腸道功能及機(jī)體免疫功能的提升引起的。
發(fā)酵中草藥主要通過改善腸道組織形態(tài)、提高消化酶的產(chǎn)生、調(diào)節(jié)腸道微生物組成等促進(jìn)畜禽腸道及機(jī)體健康。給感染豬流行性腹瀉病毒(PEDV)的仔豬灌服白頭翁散煎劑發(fā)酵液,可以緩解PEDV感染造成的仔豬腸道絨毛高度顯著下降,隱窩深度增高,明顯改善患病仔豬的腸道形態(tài),促進(jìn)仔豬腸道健康[42]。在斷奶仔豬日糧中添加發(fā)酵中草藥,可以觀察到仔豬胰凝乳蛋白酶、α-淀粉酶和脂肪酶活性都有顯著升高,空腸絨毛高度和隱窩深度均增加,腸道消化酶的分泌也有所增加,從而促進(jìn)了仔豬腸道健康[43]。在感染沙門菌的烏骨肉雛雞日糧中添加發(fā)酵中草藥進(jìn)行治療,發(fā)現(xiàn)發(fā)酵中草藥能夠降低感染雛雞腸道內(nèi)的腸炎沙門菌豐度,增加腸道中有益菌群的豐度,減輕感染對(duì)雛雞腸組織結(jié)構(gòu)的損傷[44]。在沙門菌感染肉仔雞日糧中添加發(fā)酵中草藥,相比于未發(fā)酵組,更能增加盲腸乳酸桿菌數(shù)量,降低大腸桿菌和沙門菌數(shù)量,并能顯著降低腫瘤壞死因子-α(TNF-α)和干擾素-γ(IFN-γ)的mRNA表達(dá)水平,這表明中草藥發(fā)酵后更能有效調(diào)控沙門菌感染肉雛雞腸道菌群,從而增強(qiáng)雛雞的免疫能力[45]。在28日齡健康三元雜交斷奶仔豬日糧中添加經(jīng)發(fā)酵的中草藥復(fù)方制劑,可顯著提高斷奶仔豬腸道中的糞便乳酸桿菌和雙歧桿菌數(shù)量,同時(shí)降低糞便大腸桿菌的數(shù)量,這表明發(fā)酵中草藥能夠調(diào)節(jié)斷奶仔豬菌群結(jié)構(gòu),提高有益菌的豐度,從而改善其腸道健康狀況[46]。
有研究表明,發(fā)酵中草藥中的多糖類、生物堿類和有機(jī)酸類等活性成分能促進(jìn)機(jī)體免疫器官的生長發(fā)育,激發(fā)非特異性免疫反應(yīng),進(jìn)而增強(qiáng)動(dòng)物的免疫功能。在艾維茵肉雞日糧中添加發(fā)酵的復(fù)方中草藥制劑以及在817肉雞飲水中添加經(jīng)乳酸菌發(fā)酵的中草藥復(fù)合制劑,均能夠使脾臟指數(shù)、免疫器官指數(shù)顯著提高[15,47]。將發(fā)酵中草藥制劑添加至羅斯308肉雞日糧中,發(fā)現(xiàn)白細(xì)胞介素-1β(IL-1β)、白細(xì)胞介素-6(IL-6)、免疫球蛋白G(IgG)含量及超氧化物歧化酶(SOD)、過氧化氫酶(CAT)及谷胱甘肽過氧化物酶(GSH-Px)活性均顯著增加[48]。將經(jīng)過發(fā)酵的復(fù)方中草藥制劑添加到AA肉雞的飼料中時(shí),觀察到免疫應(yīng)激肉雞的白細(xì)胞數(shù)量、谷草轉(zhuǎn)氨酶(AST)及肌酸激酶(CK)血清酶活性顯著下降[49]。在黃羽肉雞飼糧中添加發(fā)酵中草藥,并在75日齡時(shí)注射脂多糖(LPS)應(yīng)激處理3 h后屠宰取樣,發(fā)現(xiàn)血清 IL-1β、白細(xì)胞介素-2(IL-2)和TNF-α含量顯著降低,血清白細(xì)胞介素-10(IL-10)和免疫球蛋白A(IgA)、免疫球蛋白M(IgM)、免疫球蛋白Y(IgY)含量顯著提高[50]。給新西蘭兔飼喂添加了發(fā)酵黃艾散的日糧,發(fā)現(xiàn)肉兔血清IgG和IgM含量顯著提高[51]。將經(jīng)過發(fā)酵的中草藥湯添加到育肥豬的日糧中,發(fā)現(xiàn)育肥豬血清中的IgM和補(bǔ)體C4含量顯著升高[30]。在灘羊日糧中飼喂發(fā)酵后的枸杞渣,可顯著提高血清中IgA、IgM、IgG、球蛋白(GLB)的水平,但與未發(fā)酵組差異不顯著[52]。進(jìn)一步研究表明,發(fā)酵后的枸杞渣相比未發(fā)酵枸杞渣能夠明顯提高趨化因子的表達(dá)和激活免疫相關(guān)信號(hào)通路,如IL-17信號(hào)通路、NOD樣受體信號(hào)通路等,表明發(fā)酵枸杞渣能夠改善綿羊免疫功能,從而促進(jìn)綿羊的健康狀況[52]。以上結(jié)果均表明,發(fā)酵中草藥可能通過其活性成分對(duì)畜禽免疫功能的提高,從而發(fā)揮對(duì)機(jī)體健康的促進(jìn)作用。
2.3 對(duì)畜產(chǎn)品品質(zhì)的影響
將發(fā)酵中草藥飼料添加劑添加到畜禽飼料里,可以有效提升畜產(chǎn)品的品質(zhì),并改善畜產(chǎn)品中的健康指標(biāo)。肉品質(zhì)方面,在松遼黑豬日糧中添加植物乳桿菌和酶制劑發(fā)酵的中草藥副產(chǎn)物,能夠降低平均背膘厚,提高熟肉率和油酸含量,從而改善豬肉品質(zhì),提高其營養(yǎng)價(jià)值[53]。在蒙山黑山羊的日糧中添加發(fā)酵的中草藥復(fù)方組,通過測(cè)定山羊左側(cè)眼肌、右側(cè)眼肌、肱二頭肌發(fā)現(xiàn)其剪切力和失水率均顯著降低,熟肉率、粗蛋白、粗脂肪和pH24h顯著升高,表明發(fā)酵中草藥對(duì)山羊的肉品質(zhì)有明顯改善作用[54]。在西門塔爾?!镣廃S牛雜交F1代育肥牛日糧中添加發(fā)酵的中草藥制劑,牛肉的系水率、剪切力、大理石評(píng)分顯著改善,粗蛋白含量顯著升高,對(duì)牛肉的品質(zhì)有促進(jìn)作用[55]。乳品質(zhì)方面,在飼糧中添加18種中草藥組成的發(fā)酵中草藥以及發(fā)酵的復(fù)方中草藥制劑飼喂泌乳荷斯坦奶牛,均發(fā)現(xiàn)乳脂及蛋白質(zhì)含量顯著升高,這表明發(fā)酵中草藥對(duì)奶牛的乳品質(zhì)有促進(jìn)作用[37,56]。蛋品質(zhì)方面,將酵母菌發(fā)酵的黃芪、當(dāng)歸、白術(shù)等中草藥飼料添加劑添加到羅曼粉蛋雞的日糧中,發(fā)現(xiàn)蛋雞的沙殼蛋率降低,蛋白高度與哈夫單位均顯著增加[57]。在孫雨豪[58]的研究中,將發(fā)酵中草藥添加到蛋雞飼料中,也發(fā)現(xiàn)蛋殼厚度、強(qiáng)度及哈夫單位均顯著增加,這意味著發(fā)酵中草藥對(duì)蛋雞產(chǎn)品的品質(zhì)起到了改善作用。
隨著人們生活水平的提高,對(duì)畜產(chǎn)品健康指標(biāo)的關(guān)注度也越來越高,包括畜產(chǎn)品中膽固醇水平與不飽和脂肪酸水平等。總膽固醇(TC)是人類進(jìn)行正常生命活動(dòng)不能缺少的要素,居民主要通過雞蛋獲取膳食膽固醇[59],但雞蛋中所含的膽固醇指數(shù)很高,如果攝入過多會(huì)使人體內(nèi)膽固醇指數(shù)過高,導(dǎo)致高膽固醇血癥的發(fā)生[60]。通過在京紅蛋雞的日糧中添加發(fā)酵的中草藥,觀察發(fā)現(xiàn)雞蛋中TC含量顯著降低[61-62]。將發(fā)酵后的杜仲葉粉飼喂給蛋雞,觀察到蛋雞血液中TC、甘油三酯、低密度脂蛋白的含量均顯著降低,高密度脂蛋白的含量顯著增加[63]。以上研究均表明,發(fā)酵中草藥能夠通過降低蛋的膽固醇水平,提高蛋品質(zhì)。脂肪酸是乳脂起到營養(yǎng)作用的關(guān)鍵因素,其中ω-3多不飽和脂肪酸(n-3 PUFA)作為人體必需營養(yǎng)素的重要組成部分對(duì)人體健康至關(guān)重要,它能夠使血清中甘油三酯和低密度脂蛋白的膽固醇含量顯著降低[64]。反芻動(dòng)物在生產(chǎn)中添加中草藥飼料添加劑能夠?qū)θ楫a(chǎn)品中不飽和脂肪酸含量進(jìn)行改善,提高奶制品的營養(yǎng)價(jià)值,從而使乳制品具有更多營養(yǎng)和保健功效。在荷斯坦奶牛飼糧中添加中草藥,發(fā)現(xiàn)生乳和血液中各n-3 PUFA和總n-3 PUFA水平增加,生乳中ω-6多不飽和脂肪酸(n-6 PUFA)與n-3 PUFA的比例降低[65]。李寧等[66]在荷斯坦奶牛日糧中添加中草藥,觀察到乳中α-亞麻酸(ALA)和n-3 PUFA的含量均顯著升高,還具有降低乳蛋白率和乳脂率的趨勢(shì)。雖然沒有發(fā)酵中草藥在反芻動(dòng)物乳中關(guān)于n-3 PUFA的研究,但以上結(jié)果提示發(fā)酵中草藥對(duì)反芻動(dòng)物乳中脂肪酸含量很可能有提高作用。
3 發(fā)酵中草藥飼料添加劑使用中存在的問題
3.1 標(biāo)準(zhǔn)化生產(chǎn)體系不健全
如今,微生物發(fā)酵中草藥制劑的研究雖然逐步深入,但仍然有很大的探索空間。中草藥由于內(nèi)含多種活性成分,作用于不同靶點(diǎn)、產(chǎn)生多種效應(yīng)等原因,使其藥代動(dòng)力學(xué)在研究上變得復(fù)雜多樣[67]。因此,在中草藥發(fā)酵研究過程中,仍存在著一系列困難和挑戰(zhàn)。首先,中草藥自身體系的不確定性使得研究人員難以準(zhǔn)確預(yù)測(cè)發(fā)酵過程中的變化和效果。另外,原料中成分復(fù)雜,其中包含了許多活性物質(zhì)和化合物,增加了研究的復(fù)雜性和難度。此外,在發(fā)酵過程中不同微生物的作用和微生物轉(zhuǎn)化之間的關(guān)系并未研究明確,這使得研究人員對(duì)于中草藥中具體哪些成分起到了主導(dǎo)作用也不得而知[68]。因此,運(yùn)用先進(jìn)的技術(shù)和方法,探究中草藥藥代動(dòng)力學(xué),理解其藥效和作用機(jī)制,這對(duì)于發(fā)現(xiàn)新的有效成分和開發(fā)更安全有效的中草藥制劑,為畜禽生產(chǎn)上的應(yīng)用能夠提供更好的選擇。此外需開展藥用微生物菌種選育研究,運(yùn)用現(xiàn)代發(fā)酵技術(shù),分離并篩選出具有優(yōu)勢(shì)特性的微生物菌種,以適用于不同單味中草藥和復(fù)方中草藥的高效發(fā)酵[69]。發(fā)酵中草藥飼料添加劑標(biāo)準(zhǔn)化和生產(chǎn)體系的不健全,也使得發(fā)酵中草藥在畜禽生產(chǎn)中的應(yīng)用比例還較低。為了確保微生物發(fā)酵中草藥制劑在生產(chǎn)上的穩(wěn)定性,還需要加強(qiáng)對(duì)發(fā)酵中草藥質(zhì)量標(biāo)準(zhǔn)的研究,為相關(guān)研究和開發(fā)提供規(guī)范性的指導(dǎo),從而進(jìn)一步促進(jìn)中草藥產(chǎn)業(yè)的發(fā)展和創(chuàng)新。標(biāo)準(zhǔn)化生產(chǎn)體系的完善,對(duì)于逐步建立起新型的高效發(fā)酵中草藥飼料添加劑體系至關(guān)重要。
3.2 產(chǎn)業(yè)鏈發(fā)展不完善
我國禁止在飼料端添加抗生素后,各畜禽養(yǎng)殖企業(yè)和飼料生產(chǎn)企業(yè)一直在尋找可以替代抗生素的飼料添加劑。隨著綠色、新型飼料添加劑的需求越來越大,中草藥飼料添加劑逐漸被認(rèn)為是最具潛力的抗生素替代[70],這為發(fā)酵中草藥在畜牧行業(yè)中的發(fā)展提供了重要的機(jī)遇。但發(fā)酵中草藥飼料添加劑的產(chǎn)業(yè)還并未形成完善的研發(fā)、生產(chǎn)、應(yīng)用以及更進(jìn)一步的高水平中草藥發(fā)展產(chǎn)業(yè)鏈,也存在有科技創(chuàng)新成果轉(zhuǎn)化乏力、企業(yè)間分工合作乏力、中小企業(yè)資金困難等問題。隨著國家大力發(fā)展推進(jìn)中草藥獸藥和中草藥飼料添加劑等生態(tài)養(yǎng)殖投入品助力養(yǎng)殖產(chǎn)業(yè)生態(tài)化變革的同時(shí),有關(guān)部門應(yīng)積極發(fā)揮主導(dǎo)作用,推動(dòng)相關(guān)產(chǎn)業(yè)鏈的大力發(fā)展,助力形成一條健全完善的新型市場(chǎng)產(chǎn)業(yè)鏈,對(duì)新興企業(yè)發(fā)展提供資金支持服務(wù),政策普惠,專家協(xié)助等。助農(nóng)部門帶動(dòng)形成龍頭企業(yè),協(xié)助中小企業(yè)發(fā)展,并鼓勵(lì)龍頭企業(yè)發(fā)揮聯(lián)農(nóng)帶農(nóng)機(jī)制,為養(yǎng)殖場(chǎng)提供個(gè)性化服務(wù),幫助養(yǎng)殖場(chǎng)合理化、高效化利用發(fā)酵中草藥添加劑,提高發(fā)酵中草藥的使用普及率[71],進(jìn)一步推動(dòng)發(fā)酵中草藥產(chǎn)業(yè)鏈完善。
3.3 養(yǎng)殖戶接受度不高
發(fā)酵中草藥是一種理想的抗生素替代品,在未來畜禽養(yǎng)殖行業(yè)具有巨大應(yīng)用潛力[72]。大量研究證實(shí),在畜禽生產(chǎn)中添加發(fā)酵中草藥飼料添加劑對(duì)動(dòng)物生產(chǎn)性能和提高抗病力都具有改善作用。在養(yǎng)殖生產(chǎn)過程中通過對(duì)飼料配比環(huán)節(jié)進(jìn)行優(yōu)化,從而降低畜禽飼養(yǎng)成本、有效提高養(yǎng)殖經(jīng)濟(jì)效益,獲得最大生產(chǎn)收益[73]。但大多數(shù)養(yǎng)殖戶對(duì)于新型飼料添加劑的了解非常少,對(duì)傳統(tǒng)養(yǎng)殖生產(chǎn)依賴度高。部分農(nóng)戶對(duì)于發(fā)酵中草藥類飼料添加劑的認(rèn)知還很初步,例如認(rèn)為原料貴、見效慢、品種繁多等,這都需要相關(guān)部門對(duì)養(yǎng)殖場(chǎng)進(jìn)行知識(shí)普及和服務(wù),對(duì)發(fā)酵中草藥飼料添加劑生產(chǎn)和應(yīng)用進(jìn)行科學(xué)普及,增強(qiáng)養(yǎng)殖戶對(duì)中草藥飼料添加劑的認(rèn)可程度,鼓勵(lì)養(yǎng)殖戶在畜禽生產(chǎn)中進(jìn)行使用。
4 小結(jié)與展望
隨著畜禽飼料中抗生素類添加劑的全面禁用,畜禽養(yǎng)殖生產(chǎn)中對(duì)替代抗生素的綠色、安全、高效飼料添加劑的需求日益迫切,而中草藥和發(fā)酵中草藥飼料添加劑由于其安全、有效且毒性小的優(yōu)點(diǎn),在畜禽養(yǎng)殖中迎來了重要機(jī)遇。發(fā)酵中草藥技術(shù)結(jié)合現(xiàn)代生物學(xué)技術(shù),微生物發(fā)酵技術(shù)與中草藥藥理學(xué)對(duì)中草藥成分進(jìn)行生物學(xué)轉(zhuǎn)化,提高藥效成分含量、產(chǎn)生新的活性物質(zhì)、降低毒副作用等,從而在動(dòng)物生產(chǎn)中發(fā)揮促進(jìn)作用。發(fā)酵處理后的中草藥對(duì)畜禽的生產(chǎn)性能、健康狀況和畜產(chǎn)品品質(zhì)等均具有明顯的改善作用,在生產(chǎn)中作為效果良好的綠色飼料添加劑,具有較大的應(yīng)用潛力。盡管發(fā)酵中草藥飼料添加劑在畜禽生產(chǎn)上的應(yīng)用效果已被證實(shí),但具體的作用機(jī)制仍需深入挖掘。此外,還需進(jìn)一步尋求高效和靶向性的微生物發(fā)酵菌種,以提高發(fā)酵中草藥的質(zhì)量和生產(chǎn)效率。在此基礎(chǔ)上,開展發(fā)酵中草藥飼料添加劑的科學(xué)評(píng)估與規(guī)范利用,必將推動(dòng)發(fā)酵中草藥飼料添加劑在動(dòng)物生產(chǎn)中的科學(xué)利用和可持續(xù)發(fā)展。
