大環(huán)內(nèi)酯類抗生素是一類分子中含有14~16元大環(huán)內(nèi)酯結(jié)構(gòu)的抗生素,因其抗菌譜廣、組織分布廣泛、副作用較少且價格低廉,在醫(yī)藥領(lǐng)域和農(nóng)藥領(lǐng)域都有著廣泛的應(yīng)用[1-3]。第一代大環(huán)內(nèi)酯類抗生素以紅霉素為代表(圖1),主要包括紅霉素及其衍生物,如琥乙紅霉素、硬脂酸紅霉素等。該代抗生素抗菌譜廣、組織分布廣泛、藥動學(xué)性質(zhì)良好,但在胃酸等酸性條件下不穩(wěn)定,易降解失活,并導(dǎo)致胃腸道不良反應(yīng)[4]。第二代大環(huán)內(nèi)酯類抗生素包括克拉霉素、氟紅霉素、阿奇霉素等(圖1)。分別針對紅霉素的C-6、C-8和C-9等不同位點進行結(jié)構(gòu)改造,極大地增強了酸穩(wěn)定性、延長了半衰期、減緩了胃腸道不良反應(yīng)[5]。 然而,隨著耐藥菌的不斷出現(xiàn),對新型抗生素的需求更加迫切。1977年,Allen等[6]發(fā)現(xiàn),C-3上的克拉定糖并非大環(huán)內(nèi)酯類抗生素發(fā)揮抗菌活性的必需基團,相反的,它還與誘導(dǎo)細(xì)菌產(chǎn)生耐藥性相關(guān),因此圍繞C-3脫克拉定糖后進行結(jié)構(gòu)修飾的第三代大環(huán)內(nèi)酯類抗生素(主要包括酰內(nèi)酯與酮內(nèi)酯)應(yīng)運而生。泰利霉素是目前唯一上市的第三代大環(huán)內(nèi)酯類抗生素,它不僅拓寬了大環(huán)內(nèi)酯類抗生素的抗菌譜,而且還降低了細(xì)菌耐藥性,大大增強了抗菌活性。但由于其嚴(yán)重的肝毒性,美國食品藥品監(jiān)督管理局(FDA)不得不對其適應(yīng)癥進行限制[7-9]。
基于大環(huán)內(nèi)酯類抗生素的構(gòu)效關(guān)系,本文設(shè)計兩類大環(huán)內(nèi)酯類衍生物:①以紅霉素為原料,先合成氟紅霉素,并去除C-3位克拉定糖,以期降低耐藥性,對產(chǎn)生的C-3位羥基進行酯化反應(yīng)彌補可能的抗菌活性的降低,同時C-11、C-12環(huán)合成碳酸酯增強抗菌活性[10],合成了衍生物5a~5k(圖2);②以克拉霉素為原料去除C-3的克拉定糖,氧化為羰基,降低耐藥性并提高對erm基因介導(dǎo)的耐藥菌的抑制活性[11],并對C-11、C-12位進行環(huán)合衍生化[12-13],獲得衍生物12a~12f(圖2)。
AscendTM400型核磁共振儀(德國Bruker公司),MAT-95XP型高分辨質(zhì)譜儀(德國Bruker公司),F(xiàn)LUOstar?OPTIMA型多功能酶標(biāo)儀(德國BMG Labtecg公司)。
1-氯甲基-4-氟-1,4-重氮化二環(huán)2.2.2辛烷雙(四氟硼酸鹽)(selectflour)采購自上海鷹谷信息科技有限公司,紅霉素和克拉霉素由東陽光研究院提供,其它試劑除特殊說明外均為市售分析純試劑。
紅霉素(100.1g, 136.4 mmol)和1-氯甲基-4-氟-1,4-重氮化二環(huán)2.2.2辛烷雙(四氟硼酸鹽)(50.0g, 141.1mmol)溶解在400mL冰醋酸中,室溫下攪拌,TLC監(jiān)控反應(yīng),6h后基本反應(yīng)完全。反應(yīng)液用6mol/L的氫氧化鈉溶液調(diào)節(jié)pH=10,二氯甲烷萃取,有機相依次用飽和碳酸氫鈉溶液、飽和食鹽水洗滌,無水硫酸鈉干燥,過濾后減壓濃縮得淡黃色固體,乙醇重結(jié)晶,得白色固體1(70.0g,收率68 %)。1H NMR(400MHz, DMSO-d6) δ(ppm): 4.81(d, J=5.8Hz, 1H), 4.77(dd, J=11.2, 2.4Hz, 1H), 4.72(d, J=4.3Hz, 1H), 4.39(s, 1H), 4.26~4.21(m, 2H), 4.18(d, J=7.3Hz, 1H), 4.08(s, 1H), 4.05~3.98(m, 3H), 3.54~3.48(m, 1H), 3.44(dd, J=7.0, 5.1Hz, 1H), 3.19(s, 3H), 3.14~3.08(m, 1H), 2.94~2.88(m, 1H), 2.87~2.85(m, 1H), 2.85~2.80(m, 1H), 2.71(dd, J=7.2, 4.9Hz, 1H), 2.47~2.42(m, 1H), 2.30~2.25(m, 1H), 2.22(s, 6H), 2.03~1.97(m, 1H), 1.94~1.89(m, 1H), 1.80~1.72(m, 1H), 1.66~1.60(m, 1H), 1.56(s, 3H), 1.48~1.45(m, 1H), 1.40(s, 3H), 1.35(s, 1H), 1.18~1.15(m, 3H), 1.14(m, 1H), 1.12(s, 3H), 1.10(s, 3H), 1.10 ~1.08(m, 6H), 1.07~1.05(m, 3H), 1.04~1.02(m, 3H), 0.78(t, J=7.3Hz, 3H)。

化合物1(70.0g, 93.0mmol)溶解在1200mL稀鹽酸(pH=1.5)中,室溫攪拌反應(yīng),TLC監(jiān)控,24h后原料基本反應(yīng)完全。反應(yīng)液用氨水調(diào)節(jié)至pH=9。乙酸乙酯萃取,有機相依次用飽和碳酸氫鈉溶液、飽和食鹽水洗滌,無水硫酸鈉干燥。過濾,濾液減壓濃縮得白色固體,柱層析(V(二氯甲烷)/V(甲醇)=20:1)得白色固體2(40.0g,收率72%)。1H NMR(400MHz, DMSO-d6) δ(ppm): 5.12(s, 1H), 5.10(s, 1H), 4.48(d, J=7.3Hz, 1H), 4.12(s, 1H), 4.04(s, 1H), 3.69(d, J=10.6Hz, 2H), 3.56(s, 1H), 3.47~3.38(m, 1H), 3.20(dd, J=9.0, 8.3Hz, 1H), 3.09~2.99(m, 2H), 2.83(s, 3H), 2.22(s, 6H), 1.99(s, 1H), 1.98~1.90(m, 1H), 1.88~1.77(m, 2H), 1.75(s, 3H), 1.61(d, J=12.3Hz, 1H), 1.43~1.30(m, 2H), 1.19(s, 3H), 1.13(d, J=6.0Hz, 3H), 1.10~1.03(m, 9H), 1.01(d, J=6.5Hz, 3H), 0.73(t, J=7.1Hz, 3H)。
化合物2(40.0g,67.3mmol)溶解在250mL四氫呋喃中,0℃下加入苯甲酸酐(18.5g, 81.8mmol),攪拌溶解后在0.5h內(nèi)緩慢滴加三乙胺9.50mL,加入DMAP(0.10g, 0.81mmol),攪拌0.5h后升至室溫,TLC監(jiān)控反應(yīng),48h后加入飽和碳酸氫鈉溶液100mL,繼續(xù)攪拌1h,靜置分層,有機層依次用飽和碳酸氫鈉溶液和飽和食鹽水洗滌,無水硫酸鈉干燥。過濾,濾液減壓濃縮得白色固體,柱層析(V(二氯甲烷)/V(甲醇)=40:1)得白色固體3(42.0g,收率89 %)。1H NMR(400MHz, DMSO-d6) δ(ppm): 7.97(d, J=7.1Hz, 2H), 7.67~7.60(m, 1H), 7.53(t, J=7.6Hz, 2H), 5.25(d, J=6.8Hz, 1H), 5.06(dd, J=11.1, 2.3Hz, 1H), 4.86~4.81(m, 2H), 4.46(s, 1H), 4.08(s, 1H), 3.77(s, 1H), 3.58~3.48(m, 1H), 3.30(d, J=3.0Hz, 1H), 3.23~3.14(m, 1H), 2.88~2.78(m, 2H), 2.42~2.35(m, 1H), 2.16(s, 6H), 2.04~1.99(m, 1H), 1.96~1.87(m, 1H), 1.77(dd, J=14.4, 6.7Hz, 2H), 1.68(d, J=15.3Hz, 1H), 1.48~1.37(m, 4H), 1.35~1.26(m, 5H), 1.20~1.16(m, 3H), 1.06(d, J=6.6Hz, 3H), 1.02(d, J=6.7Hz, 3H), 0.87(s, 3H), 0.69(t, J=7.4Hz, 3H), 0.61(d, J=7.4Hz, 3H)。
化合物3(42.0g,60.3mmol)溶解在250mL二氯甲烷中,在0℃下加入吡啶2.1mL,緩慢滴加三光氣(21.5g, 72.4mmol)的二氯甲烷溶液(50mL),緩慢滴加三乙胺30mL。繼續(xù)攪拌5.5h,向反應(yīng)瓶中加入少量的碎冰塊猝滅反應(yīng)。有機相依次用飽和碳酸氫鈉溶液、飽和食鹽水洗滌,無水硫酸鈉干燥。過濾,濾液減壓濃縮得黃色固體,柱層析(V(石油醚)/V(乙酸乙酯)=1:1)得淡黃色固體4(35.0g,收率80%)。1H NMR(400MHz, CDCl3) δ(ppm): 8.09(d, J=7.8Hz, 2H), 7.68(t, J=7.5Hz, 1H), 7.51(t, J=7.3Hz, 2H), 5.34~5.24(m, 1H), 4.90(d, J=9.3Hz, 1H), 4.86(s, 1H), 4.74(d, J=6.8Hz, 1H), 3.86~3.68(m, 2H), 3.63(s, 1H), 3.24(s, 1H), 2.79(dd, J=25.1, 3.8Hz, 6H), 2.60(t, J=13.4Hz, 1H), 2.41~2.30(m, 1H), 2.23(dd, J=23.0, 13.8Hz, 1H), 2.05(s, 1H), 1.99~1.91(m, 1H), 1.88~1.76(m, 2H), 1.76~1.69(m, 1H), 1.68~1.59(m, 6H), 1.57(s, 3H), 1.46(s, 3H), 1.36(d, J=5.3Hz, 3H), 1.12(d, J=6.6Hz, 3H), 0.88~0.79(m, 6H), 0.76(d, J=7.0Hz, 3H)。
化合物4(1.38mmol)溶解在二氯甲烷中,0℃下加入有機酸(1.7mmol)、EDC·HCl(1.7mmol)和DMAP(0.14mmol),繼續(xù)攪拌0.5h后升至室溫,反應(yīng)4d后反應(yīng)液用2mol/L的氫氧化鈉溶液調(diào)節(jié)pH=8。有機相依次用飽和氯化銨溶液、飽和食鹽水洗滌,無水硫酸鈉干燥。過濾,減壓濃縮除去溶劑,柱層析(V(石油醚)/V(乙酸乙酯)=1:1)得中間體。將上述中間體溶解在適量甲醇中,回流10h,減壓濃縮除去溶劑,柱層析(V(二氯甲烷)/V(甲醇)=30:1)得目標(biāo)產(chǎn)物5a~5k。

采用微量稀釋法測定化合物的最小抑菌濃度(MIC)。在滅菌的96孔板中加入LB培養(yǎng)液(每孔100μL)。分別設(shè)置陽性對照組(紅霉素A、克拉霉素、氟紅霉素)、空白對照組、溶劑對照組(0.4mL DMSO)。在剩余的孔中從低濃度到高濃度依次加入濃度為50mmol溶于DMSO的待測化合物溶液(重復(fù)3孔),使各孔中化合物的濃度分別為0.39、0.78、1.56、3.13、6.25、12.5、25、50、100和200μmol/L。然后在每孔中加入當(dāng)日在LB培養(yǎng)液新鮮生長、吸光度為0.6~1.0的菌液5μL。微孔板加蓋并用膠紙密封,使用多功能酶標(biāo)儀于37℃孵育18h,并使96孔板在7mm距離上下震蕩,每10min靜止并在600nm讀取吸光度,計算最小抑菌濃度。
采用微量稀釋法測定了本文獲得的17個化合物對B.subtilis 168、S.aureus USA 300、E.coli DH5a、P.aeruginosa PAO1、A.baumannii ATCC19606的體外抑菌活性,測試結(jié)果如表1所示。
氟紅霉素衍生物中,5a和5b表現(xiàn)出了較好的抗菌活性。這兩個化合物對B.subtilis 168和A.baumannii ATCC19606的活性均為氟紅霉素的4倍??死顾匮苌镏校?2d、12e和12f均表現(xiàn)出了較好的抗菌活性,其中12d的效果最為顯著,對E.coli DH5α的抑菌活性是克拉霉素的8倍,對P.aeruginosa PAO1的抑菌活性是克拉霉素的4倍。
(1)氟紅霉素衍生物C-3上的不同側(cè)鏈對化合物的抗菌活性影響較明顯。5b的抑菌活性略強于5a,提示在同為對硝基取代苯環(huán)的情況下,芳基丙酸取代較芳基乙酸取代可能更有利于提高化合物的抑菌活性。此外,C-3同為鹵代芳基乙酸取代時,苯環(huán)上鹵代的位置對抑菌活性影響不顯著,如分別為o-氟苯乙酸、m-氟苯乙酸及p-氟苯乙酸的化合物5c、5d、5e的抑菌活性并無顯著差異;(2)克拉霉素衍生物11, 12位氨基甲酸酯環(huán)上N取代側(cè)鏈對化合物的抗菌活性有較大影響。當(dāng)側(cè)鏈為脂肪直鏈時,衍生物有一定的抗菌活性,且脂肪鏈越長抑菌活性越強。C原子數(shù)目n=3時,化合物抑菌活性差,3<n≤7時抑菌活性增強?;衔?2d~12f抗菌活性較好,推測側(cè)鏈長度為4~5個原子、末端為脂肪雜環(huán)或芳環(huán)時衍生物有較好的體外活性。除S.aureus USA 300外,化合物12d對其余4種受測菌株的抑菌活性均強于化合物12e與12f,推測脂肪鏈長度為4~5個原子的情況下,長鏈末端為脂肪雜環(huán)取代可能更有利于提高抑菌活性。另外,化合物12e和12f對革蘭陽性菌的體外抑制活性均強于革蘭陰性菌,推測當(dāng)側(cè)鏈末端是芳環(huán)時,可能會表現(xiàn)出對革蘭陽性菌的選擇性。

本文對氟紅霉素和克拉霉素進行結(jié)構(gòu)改造,合成了17個新的大環(huán)內(nèi)酯類衍生物。通過抗菌活性測試,進一步探討了大環(huán)內(nèi)酯類抗生素的構(gòu)效關(guān)系,為大環(huán)內(nèi)酯類抗生素的后續(xù)研究提供了參考。
