[摘要] 目的 探討唑來(lái)膦酸對(duì)去勢(shì)大鼠骨折后骨痂和骨微結(jié)構(gòu)的影響。 方法 將45只SD雌性大鼠分為骨質(zhì)疏松組(30只)和正常對(duì)照組(15只),骨質(zhì)疏松組雌性大鼠被切除雙側(cè)卵巢制作骨質(zhì)疏松動(dòng)物模型,正常對(duì)照組大鼠僅給予假手術(shù);骨質(zhì)疏松模型建模成功后,將骨質(zhì)疏松組30只大鼠再次隨機(jī)分為生理鹽水組(15只)和唑來(lái)膦酸組(15只);對(duì)兩組大鼠制作股骨骨折模型,并分別給予生理鹽水和唑來(lái)膦酸進(jìn)行干預(yù);在藥物干預(yù)4周后,比較生理鹽水組和唑來(lái)膦酸組骨質(zhì)疏松大鼠股骨骨痂和骨微結(jié)構(gòu)情況。 結(jié)果 卵巢切除術(shù)后8周,正常對(duì)照組和骨質(zhì)疏松組大鼠股骨的骨密度分別為(0.197±0.028)g/cm2和(0.128±0.037)g/cm2,差異有統(tǒng)計(jì)學(xué)意義(P < 0.05);在藥物干預(yù)4周后,兩組骨質(zhì)疏松大鼠股骨均有骨痂形成,而唑來(lái)膦酸組大鼠骨痂明顯大于生理鹽水組,且骨折線較為模糊;與生理鹽水組大鼠比較,唑來(lái)膦酸組大鼠骨小梁間隙較小,排列也較為整齊致密,更接近正常對(duì)照組大鼠。 結(jié)論 唑來(lái)膦酸可促進(jìn)骨質(zhì)疏松大鼠骨折后骨痂形成,并提高其骨密度。

Effects of Zoledronic Acid on callus and bone microstructure after fracture in osteoporosis rats

[Abstract] Objective To investigate the effects of Zoledronic Acid on callus and bone microstructure after fracture in osteoporosis rats. Methods 45 SD female rats were divided into osteoporosis group (30 cases) and control group (15 cases), the female rats osteoporosis group were ovariectomized to make model of osteoporosis, the rats of normal control were only given sham-operated; after the model of osteoporosis was made successfully, the 30 rats of osteoporosis group were again randomly divided into saline group (15 cases) and Zoledronic Acid group (15 cases); all rats of osteoporosis group were made femur fracture model, and been given saline and Zoledronic Acid to treatment; 4 weeks after drug intervention, the callus and bone microstructure situation of rats were observed and compared. Results At 8 weeks after ovariectomy, the BMD of femur in SD rats of osteoporosis group and control group were (0.197±0.028) g/cm2 and (0.128±0.037) g/cm2 kespectively, the difference was significant (P < 0.05); after 4 weeks of drug intervention, all rats of two groups had osteoporosis femur bone callus formation, and callus in rats of Zoledronic Acid group was significantly greater than that of the saline group, and the fracture line was more blurred; compared with the rats of saline group, the trabecular bone gap in rats of Zoledronic Acid group was more smaller, compact and tidy arrangement was relatively closer to the normal control group. Conclusion Zoledronic Acid can promote the rat osteoporotic bone fracture callus formation and increase bone density.
